麗水市HIV感染長期不進展者HLA-A、HLA-B和HLA-DRB1基因多態(tài)性的研究

【摘要】:目的從基因水平了解麗水市艾滋病病毒(HIV)感染長期不進展者(LTNPs)與典型進展者(TPs)的人類白細胞抗原(HLA)-A、HLA-B和HLA-DRB1位點等位基因頻率,比較兩類人群的差異,初步探索保護性基因和易感基因。方法應用聚合酶鏈反應-序列特異性引物技術(PCR-SSP),對麗水市42例LTNPs和48例TPs進行HLA-A、HLA-B和HLA-DRB1等位基因分型。結果共鑒定出9個HLA-A等位基因,16個HLA-B等位基因,12個HLA-DRB1等位基因。HLA-A*02頻率LTNPs組(44.05%)高于TPs組(20.83%)(P0.01),而A*11頻率則是LINPs組(20.24%)低于TPs組(42.71%)(P0.01);HLA-B*40頻率LTNPs組(17.86%)低于TPs組(28.13%)(P0.05);HLA-DRB*15頻率LTNPs組(5.95%)低于TPs組(29.17%)(P0.01);LTNPs組HLA基因座純合子頻率(15.87%)低于TPs組(33.33%)(P0.01)。結論在該人群中,HLA-A*02等位基因可能與延緩艾滋病疾病進程相關,而HLA-A*11、B*40、DRB1*15可能加速艾滋病疾病進程。含HLA基因雜合子者對疾病顯示出更好的抵御能力。
[Abstract]:Objective to study the allelic frequencies of HLA-B and HLA-DRB1 loci of human leukocyte antigen (HLA) in Lishui city of China (Lishui city) with HIV / AIDS) infection for a long time, and to compare the allelic frequencies of HLA-B and HLA-DRB1 loci between patients with chronic HIV / AIDS infection and those with typical progression (TPS), and to compare the allelic frequencies between the two populations. To explore the protective and susceptible genes. Methods Polymerase chain reaction-sequence specific primer technique (PCR-SSPN) was used to type HLA-Ag HLA-B and HLA-DRB1 alleles in 42 cases of LTNPs and 48 cases of TPs in Lishui city. Results A total of 9 HLA-A alleles, 16 HLA-B alleles and 12 HLA-DRB1 alleles were identified in LTNPs group (44.05) higher than TPs group (20.83% P 0.01), and An11 frequency was lower in LINPs group (20.24%) than TPs group (P 0.01HLA-BN 40 frequency LTNPs group (17.8686) lower than TPs group (28.13%) in LTNPs group (5.95). The homozygote frequency of HLA locus in group P 0.01 was lower than that in group TPs (P 0.01). Conclusion HLA-An02 allele may be associated with delaying the progression of HIV / AIDS in this population, while HLA-A11 / BX 40 / DRB1 / 15 may accelerate the progression of HIV / AIDS. Heterozygotes with HLA genes showed better resistance to disease.
