利福噴丁對(duì)大鼠細(xì)胞因子及巨噬細(xì)胞吞噬功能的影響

【摘要】:目的:研究利福噴丁對(duì)大鼠細(xì)胞因子IFN-γ、IL-12、IL-6及巨噬細(xì)胞吞噬功能的影響。方法:1.60只SD雄性大鼠,二月齡,體重為180±30g,隨機(jī)分為實(shí)驗(yàn)組和對(duì)照組,每組為30只,每組再隨機(jī)分為喂藥一月組(A組)、喂藥三月組(B組)、喂藥三月停喂藥一月組(以下簡稱為停藥一月組)(C組)(每組為10只)。2.實(shí)驗(yàn)組用利福噴丁灌胃,對(duì)照組用0.1%羧甲基纖維素鈉液灌胃,其余喂養(yǎng)條件相同。3.ELISA法檢測大鼠血清IFN-γ、IL-12、IL-6含量,瓊脂平板法檢測大鼠血清溶菌酶含量,硝酸鉛法顯示大鼠腹腔巨噬細(xì)胞內(nèi)酸性磷酸酶活性。IFN-γ、IL-12、IL-6、溶菌酶結(jié)果用t檢驗(yàn)統(tǒng)計(jì);酸性磷酸酶用軼和檢驗(yàn)統(tǒng)計(jì)。結(jié)果:1.實(shí)驗(yàn)組IFN-γ在喂藥三月較對(duì)照組有明顯降低,差異有統(tǒng)計(jì)學(xué)意義(P<0.05),在喂藥一月、停藥一月較對(duì)照組比較,差異無統(tǒng)計(jì)學(xué)意義;實(shí)驗(yàn)組IL-12在喂藥一月、喂藥三月、停藥一月較對(duì)照組比較,差異均無統(tǒng)計(jì)學(xué)意義;實(shí)驗(yàn)組溶菌酶在喂藥三月較對(duì)照組有明顯升高,差異有統(tǒng)計(jì)學(xué)意義(P<0.05),在喂藥一月、停藥一月較對(duì)照組比較,差異均無統(tǒng)計(jì)學(xué)意義;實(shí)驗(yàn)組酸性磷酸酶在喂藥一月、喂藥三月較對(duì)照組有明顯升高,差異均有統(tǒng)計(jì)學(xué)意義(P<0.05),在停藥一月較對(duì)照組比較,差異無統(tǒng)計(jì)學(xué)意義。2.對(duì)照組組內(nèi)比較:喂藥三月組、停藥一月組溶菌酶較喂藥一月組比較,結(jié)果有明顯降低,差異均有統(tǒng)計(jì)學(xué)意義(P<0.05);喂藥三月組、停藥一月組酸性磷酸酶較喂藥一月組比較,結(jié)果有明顯升高,差異均有統(tǒng)計(jì)學(xué)意義(P<0.05)。IFN-γ、IL-12在對(duì)照組內(nèi)比較,差異均無統(tǒng)計(jì)學(xué)意義。3.ELISA法在所有組中均未檢測出指標(biāo)IL-6,可能的原因是IL-6在大鼠體內(nèi)的含量很低,未達(dá)到ELISA檢測的最低檢測標(biāo)準(zhǔn)。結(jié)論:1.利福噴丁顯著降低大鼠血清IFN-γ的含量。利福噴丁降低大鼠血清IL-12含量,但降低未達(dá)到統(tǒng)計(jì)學(xué)差異。利福噴丁顯著提高大鼠血清溶菌酶含量,并且顯著增強(qiáng)大鼠腹腔巨噬細(xì)胞酸性磷酸酶活性。2.隨著大鼠月齡的增長,大鼠血清溶菌酶含量降低,腹腔巨噬細(xì)胞酸性磷酸酶活性增強(qiáng)。3.停藥后,大鼠血清IFN-γ、IL-12和溶菌酶含量恢復(fù),,大鼠腹腔巨噬細(xì)胞酸性磷酸酶活性恢復(fù)。
[Abstract]:Objective: to study the effect of rifapentine on cytokine IFN- 緯 -IL-12 IL-6 and macrophage phagocytosis in rats. Methods: one hundred and sixty SD male rats, aged two months, with body weight of 180 鹵30 g, were randomly divided into experimental group and control group with 30 rats in each group. Each group was randomly divided into three groups: group A (group A), group B (group B), group A (group A), group B (group B), group B (group B), group C (group C), group C (10 rats in each group). In the control group, 0.1% sodium carboxymethylcellulose was used to gavage, and the other feeding conditions were the same. 3. Elisa method was used to detect the content of serum IFN- 緯 -IL-12 and IL-6, and Agar plate method was used to detect the content of serum lysozyme in rats. Lead nitrate method showed that the activity of acidic phosphatase in peritoneal macrophages. IFN- 緯 -IL-12 and IL-6, lysozyme results were counted by t test, acid phosphatase by Yi and test statistics. Results: 1. IFN- 緯 in experimental group was significantly lower than that in control group in three months. The difference was statistically significant (P < 0.05). There was no significant difference in IL-12 between the control group and the control group in one month and one month after the drug was given, but there was no significant difference between the experimental group and the control group in one month, three months, and one month, respectively. The lysozyme of the experimental group was significantly higher than that of the control group in three months, the difference was statistically significant (P < 0.05), but there was no significant difference between the experimental group and the control group in one month, and the acid phosphatase in the experimental group was fed for one month. Compared with the control group, the difference was statistically significant (P < 0.05). There was no significant difference between the control group and the control group in one month. The comparison between the control group and the control group: the lysozyme in the three-month group and the one-month group were significantly higher than those in the one-month group. The results showed that the difference was statistically significant (P < 0.05), and the acid phosphatase level was significantly higher in the three months group than that in the control group (P < 0.05. IFN- 緯 -IL-12, P < 0.05, P < 0.05, P < 0.05, P < 0.05), and the level of IL-12 in the control group was significantly higher than that in the control group (P < 0.05, P < 0.05). There was no significant difference. 3. Elisa did not detect IL-6 in all groups, which may be due to the low content of IL-6 in rats. Conclusion: 1. Rifapentine can significantly decrease the content of IFN- 緯 in serum of rats, and rifapentine can decrease the content of IL-12 in serum of rats. But the decrease was not statistically significant. Rifapentine significantly increased the content of serum lysozyme and increased the activity of acid phosphatase in peritoneal macrophages of rats significantly. With the increase of the age of rats, the content of serum lysozyme decreased. The activity of acidic phosphatase in peritoneal macrophages was enhanced .3.After drug withdrawal, the contents of IL-12 and lysozyme in serum IFN- 緯 and lysozyme were recovered, and the activity of acid phosphatase in peritoneal macrophages was recovered.
