【摘要】：目的研究microRNA-122(mi R-122)在HBV相關(guān)性肝癌中的表達(dá)及其意義。方法分別提取47例肝癌組織(其中HBsAg陽性36例,HBsAg陰性11例)及癌旁正常組織的總miRNA,應(yīng)用實(shí)時(shí)熒光定量RTPCR技術(shù)檢測miR-122的表達(dá),分析其表達(dá)情況及其與臨床病理特征的關(guān)系。結(jié)果 HBsAg陽性肝癌組織中miR-122的表達(dá)比癌旁組織及HBsAg陰性肝癌組織明顯下調(diào),差異有統(tǒng)計(jì)學(xué)意義(P0.05),在肝癌組織中miR-122的表達(dá)量與患者性別、年齡、腫瘤大小無相關(guān)性(P0.05),但與腫瘤TNM分期及是否有脈管侵犯關(guān)系密切(P0.05)。結(jié)論 miR-122在HBV相關(guān)性肝癌組織中的表達(dá)明顯降低,提示miR-122的低表達(dá)與HBV相關(guān)性肝癌發(fā)生、發(fā)展存在密切關(guān)系,且miR-122表達(dá)水平可能成為診斷及判斷肝癌預(yù)后的新方法。
[Abstract]:Objective to study the expression and significance of microRNA-122(mi R-122 in HBV associated hepatocellular carcinoma (HCC). Methods Total miRNAs were extracted from 47 cases of HCC tissues (including 36 cases of HBsAg positive and 11 cases of negative HBsAg) and adjacent normal tissues. The expression of miR-122 was detected by real-time fluorescence quantitative RTPCR technique. Results the expression of miR-122 in HBsAg positive HCC was significantly lower than that in paracancerous tissues and HBsAg negative HCC tissues. The difference was statistically significant (P 0.05). The expression of miR-122 in HCC was correlated with the sex and age of the patients. There was no correlation between tumor size and tumor size (P 0.05), but it was closely related to tumor TNM stage and vascular invasion. Conclusion the expression of miR-122 in HBV related liver cancer is significantly decreased, suggesting that the low expression of miR-122 is closely related to the occurrence and development of HBV related hepatocellular carcinoma. The expression of miR-122 may be a new method for diagnosis and prognosis of HCC.
