【摘要】：為了快速鑒別診斷一例人絳蟲(chóng)感染病例,采用聚合酶鏈?zhǔn)椒?PCR)擴(kuò)增線粒體cox1和Cytb基因片段,對(duì)PCR擴(kuò)增的產(chǎn)物經(jīng)瓊脂糖凝膠電泳檢測(cè)和基因序列測(cè)序。分別與NCBI數(shù)據(jù)庫(kù)中帶絳蟲(chóng)cox1和Cytb基因序列進(jìn)行比對(duì),同時(shí)選用MEGA構(gòu)建系統(tǒng)進(jìn)化樹(shù)。結(jié)果:核酸序列與NCBI數(shù)據(jù)庫(kù)中牛帶絳蟲(chóng)同源性均為99%,進(jìn)化分析表明與牛帶絳蟲(chóng)的親緣性最近。表明該病例屬于牛帶絳蟲(chóng)感染。
[Abstract]:In order to quickly differentiate and diagnose a human tapeworm infection, mitochondrial cox1 and Cytb gene fragments were amplified by polymerase chain reaction (PCR). The products of PCR amplification were detected by agarose gel electrophoresis and sequenced, and compared with the cox1 and Cytb gene sequences of Taenia solium in NCBI database. At the same time, MEGA was used to construct phylogenetic tree. Results: the nucleic acid sequence was 99% homology to Taenia bovis in NCBI database. The phylogenetic analysis showed that the phylogenetic relationship between Taenia solium and Taenia bovis was the most recent, which indicated that the case belonged to Taenia bovis infection.
