神經(jīng)干細(xì)胞對去血清誘導(dǎo)PC12細(xì)胞凋亡的作用

【摘要】:腎上腺嗜鉻細(xì)胞瘤細(xì)胞(PC12cells)去血清后,其凋亡與多巴胺能神經(jīng)元的凋亡有許多共同之處,今通過研究神經(jīng)干細(xì)胞(NSCs)對去血清誘導(dǎo)的PC12細(xì)胞凋亡的作用,進(jìn)一步為NSCs移植治療神經(jīng)系統(tǒng)疾病提供相應(yīng)的實(shí)驗(yàn)和理論依據(jù)。將正常培養(yǎng)的PC12細(xì)胞與NSCs以不同的方式進(jìn)行去血清共培養(yǎng),觀察PC12細(xì)胞的形態(tài),檢測PC12細(xì)胞的活性,計(jì)算PC12細(xì)胞的存活率,同時(shí)檢測培養(yǎng)基中膠質(zhì)細(xì)胞源神經(jīng)營養(yǎng)因子(GDNF)的濃度,分析不同培養(yǎng)方式下NSCs對去血清誘導(dǎo)凋亡的PC12細(xì)胞的作用以及NSCs與去血清誘導(dǎo)凋亡的PC12細(xì)胞共培養(yǎng)后,其分泌GDNF的能力。結(jié)果表明:①去血清誘導(dǎo)PC12細(xì)胞凋亡呈時(shí)間依賴性,去血清72h后,PC12細(xì)胞存活的比率為44.25%;②NSCs培養(yǎng)基對去血清誘導(dǎo)凋亡的PC12細(xì)胞沒有明顯的保護(hù)作用;③NSCs培養(yǎng)上清及NSCs對去血清誘導(dǎo)凋亡的PC12細(xì)胞具有保護(hù)作用;④NSCs與去血清誘導(dǎo)凋亡的PC12細(xì)胞共培養(yǎng)后,分泌GDNF的能力增強(qiáng)。
[Abstract]:The apoptosis of adrenal pheochromocytoma cells (PC12cells) after serum removal has a lot in common with the apoptosis of dopaminergic neurons. The effect of neural stem cell (NSCs) on the apoptosis of PC12 cells induced by serum removal is studied in order to provide the corresponding experimental and theoretical basis for NSCs transplantation in the treatment of nervous system diseases. Normal cultured PC12 cells were co-cultured with NSCs in different ways. The morphology of PC12 cells was observed, the activity of PC12 cells was detected, the survival rate of PC12 cells was calculated, and the concentration of glial cell-derived neurotrophic factor (GDNF) in culture medium was also detected. The effect of NSCs on serum-free induced apoptosis of PC12 cells and the co-culture of NSCs with serum-free apoptotic PC12 cells were analyzed. Its ability to secrete GDNF.
