小鼠主動脈弓不同程度縮窄模型的評價與比較

【摘要】:目的:采用超聲多普勒的方法評價小鼠主動脈弓縮窄模型建立的成敗及程度,比較主動脈弓縮窄程度對心臟形態(tài)及功能影響的差異。方法:60只雄性C57BL/6小鼠隨機分成假手術(shù)組(sham組)20只,26G縮窄針主動脈弓縮窄組(TAC26G組)20只,27G縮窄針主動脈弓縮窄組(TAC27G組)20只,采用超聲多普勒的方法檢測小鼠雙側(cè)頸動脈流速,通過右側(cè)頸動脈(right carotid arteries,RCA)流速與左側(cè)頸動脈(left carotid arteries,LCA)流速之比(RCA/LCA)來評價主動脈弓縮窄術(shù)的成敗及程度;術(shù)后2周進行超聲心動圖檢測,并對各組心臟進行拍照,計算心重/體重及左室重/脛骨長度,比較主動脈弓不同縮窄程度對心肌肥大形成的嚴(yán)重程度以及心臟功能的影響。結(jié)果:與sham組相比,TAC26G組及TAC27G組RCA/LCA均顯著上升(P0.05),TAC27G組的RCA/LCA為8~10,較之TAC26G組(RCA/LCA為5~7)具有顯著性差異(P0.05)。術(shù)后2周,TAC26G組表現(xiàn)為向心性肥大,與sham組相比,舒張期室間隔厚度(IVSd)、舒張期后壁厚度(LVPWd)、射血分數(shù)(EF)、短軸縮短率(FS)均顯著上升(P0.05),左室舒張末期內(nèi)徑(LVIDd)顯著下降(P0.05);TAC27G組表現(xiàn)為離心性肥大,與sham組相比,IVSd、LVPWd、LVIDd均顯著上升(P0.05),EF及FS均顯著下降(P0.05)。術(shù)后2周,與sham組相比,TAC26G及TAC27G組心重/體重及左室重/脛骨長度均顯著上升(P0.05),TAC27G組上述兩個肥大指標(biāo)較之TAC26G組明顯增加(P0.05)。結(jié)論:采用超聲多普勒檢測RCA/LCA在實驗早期即可準(zhǔn)確判斷主動脈弓縮窄術(shù)的成敗及程度,對壓力超負荷心肌肥大及心力衰竭相關(guān)研究動物模型的選擇具有重要的指導(dǎo)作用。
[Abstract]:Aim: to evaluate the success or failure and degree of coarctation of aortic arch in mice by ultrasonic Doppler, and to compare the effects of coarctation of aortic arch on cardiac morphology and function. Methods: sixty male C57BL/6 mice were randomly divided into sham operation group (sham group, n = 20), 26G coarctation needle aortic arch coarctation group (TAC26G group, n = 20) and 27G coarctation needle aortic arch coarctation group (TAC27G group, n = 20). The bilateral carotid artery velocity in mice was measured by ultrasonic Doppler. The success or failure and degree of aortic arch constriction were evaluated by the ratio of right carotid artery (right carotid arteries,RCA velocity to left carotid artery (left carotid arteries,LCA velocity (RCA/LCA). Echocardiography was performed 2 weeks after operation, and the hearts of each group were photographed. The heart weight / body weight and left ventricular weight / tibia length were calculated. The effects of different coarctation of aortic arch on the severity of myocardial hypertrophy and cardiac function were compared.
