【摘要】： 目的研究廣西馬爾尼菲青霉菌(PM)的基因多態(tài)性,比較PM竹鼠分離株和臨床分離株的基因多態(tài)性差異。 方法采用氯化芐提取法提取基因組DNA。用微衛(wèi)星標記多態(tài)性分析法對40株竹鼠分離PM和31株臨床分離PM菌株進行基因多態(tài)性分析。 結果①40只廣西野生銀星竹鼠自然攜帶PM陽性率100%。②微衛(wèi)星標記多態(tài)性分析從71株PM中檢測到53個微衛(wèi)星類型,并具有較高區(qū)分度(D=0.990)。微衛(wèi)星Ⅰ,Ⅱ,Ⅲ分別檢測到21,10,和9個等位基因。③廣西銀星竹鼠體內寄生PM各地區(qū)間的遺傳相似系數為0.1217～0.4464之間。④廣西竹鼠PM與廣西馬爾尼菲青霉病患者(PSM)分離PM遺傳相似系數為0.8689.⑤廣西野生銀星竹鼠PM與廣西PSM患者分離PM各地區(qū)間的遺傳相似系數為0.0604～0.8418之間。 結論廣西馬爾尼菲青霉菌存在的基因多態(tài)性;廣西銀星竹鼠自然攜帶之PM存在地域上的基因多態(tài)性;引起人類PSM的病原菌可能與廣西野生竹鼠自然攜帶PM有密切的相關性。
[Abstract]:Aim to study the gene polymorphism of Penicillium marneffei (PM) in Guangxi and to compare the gene polymorphism between PM strain and clinical isolate. Methods Genomic DNA. was extracted by benzyl chloride extraction method. The genetic polymorphisms of 40 strains of PM isolated from bamboo mice and 31 strains of clinical isolates of PM were analyzed by microsatellite marker polymorphism analysis. Results 53 microsatellite types were detected from 71 strains of PM by microsatellite marker polymorphism analysis, with a high differentiation (D = 0.990).
