廣西馬爾尼菲青霉菌微衛(wèi)星多態(tài)性

【摘要】: 目的研究廣西馬爾尼菲青霉菌(PM)的基因多態(tài)性,比較PM竹鼠分離株和臨床分離株的基因多態(tài)性差異。 方法采用氯化芐提取法提取基因組DNA。用微衛(wèi)星標(biāo)記多態(tài)性分析法對40株竹鼠分離PM和31株臨床分離PM菌株進(jìn)行基因多態(tài)性分析。 結(jié)果①40只廣西野生銀星竹鼠自然攜帶PM陽性率100%。②微衛(wèi)星標(biāo)記多態(tài)性分析從71株P(guān)M中檢測到53個微衛(wèi)星類型,并具有較高區(qū)分度(D=0.990)。微衛(wèi)星Ⅰ,Ⅱ,Ⅲ分別檢測到21,10,和9個等位基因。③廣西銀星竹鼠體內(nèi)寄生PM各地區(qū)間的遺傳相似系數(shù)為0.1217~0.4464之間。④廣西竹鼠PM與廣西馬爾尼菲青霉病患者(PSM)分離PM遺傳相似系數(shù)為0.8689.⑤廣西野生銀星竹鼠PM與廣西PSM患者分離PM各地區(qū)間的遺傳相似系數(shù)為0.0604~0.8418之間。 結(jié)論廣西馬爾尼菲青霉菌存在的基因多態(tài)性;廣西銀星竹鼠自然攜帶之PM存在地域上的基因多態(tài)性;引起人類PSM的病原菌可能與廣西野生竹鼠自然攜帶PM有密切的相關(guān)性。
