A型肉毒毒素對(duì)電場(chǎng)刺激及SP引發(fā)的胃離體平滑肌收縮作用及機(jī)理研究

【摘要】: 目的:觀察A型肉毒毒素(botulinum toxin A,BTX-A)對(duì)大鼠幽門、胃竇離體平滑肌條自發(fā)性收縮和電場(chǎng)刺激(electrical field stimulation,EFS)、P物質(zhì)(substance P,SP)引發(fā)的幽門、胃竇離體平滑肌收縮的影響,并探討其機(jī)制。 方法:選取健康的Sprague-Dawley大鼠,體重200~250g,雌雄不拘。實(shí)驗(yàn)前禁食24h,飲水不限。實(shí)驗(yàn)時(shí)擊頭部致昏后取幽門、胃竇平滑肌各一條,置于37℃Krebs液的恒溫平滑肌槽中,肌槽內(nèi)持續(xù)供給95%O_2和5%CO_2的混合氣體,肌條的一端固定在肌槽底部的玻璃彎鉤上,另一端固定在張力傳感器上,肌條被懸于兩個(gè)銀電極之間,肌條長(zhǎng)軸與電極長(zhǎng)軸平行,電極與電刺激器相連。肌條在1g的前負(fù)荷下溫育,隨機(jī)分為對(duì)照組(control,n=12)、電場(chǎng)刺激組(electrical field stimulation,EFS,n=12)、EFS+BTX-A組(n=12)、BTX-A組(n=12)、SP組(n=12)、SP+BTX-A組(n=12)、SP+NK_1受體拮抗劑([d-arg~1,d-phe~5,d-trp~(7.9),leu~(11)]-substance P)組(n=12)。在自發(fā)性收縮的條件下,分別加入BTX-A(10 U/ml)、SP(1μM/L)、NK_1受體拮抗劑(1μM/L)及EFS(頻率16Hz,電壓35V,波寬0.5ms持續(xù)時(shí)間60-s),Biolap 420E生物機(jī)能實(shí)驗(yàn)系統(tǒng)記錄胃肌條收縮情況。 結(jié)果:1.EFS引發(fā)幽門平滑肌張力增強(qiáng)(P<0.01)和振幅增強(qiáng)(P<0.05),EFS引發(fā)胃竇平滑肌張力及振幅增強(qiáng)(P<0.01);2.BTX-A抑制EFS引發(fā)的幽門、胃竇平滑肌張力和振幅(P<0.01);3.BTX-A導(dǎo)致幽門、胃竇平滑肌張力和振幅降低(P<0.01);4.SP引發(fā)幽門平滑肌振幅增強(qiáng)(P<0.05),張力無(wú)顯著性差異(P>0.05),SP引發(fā)胃竇平滑肌張力增強(qiáng)(P<0.01)和振幅增強(qiáng)(P<0.01);5.BTX-A抑制SP引發(fā)的幽門平滑肌振幅(P<0.01),不抑制SP引發(fā)的幽門平滑肌張力(P>0.05),BTX-A抑制SP引發(fā)的胃竇平滑肌張力和振幅(P<0.01);6.NK_1受體拮抗劑抑制SP引發(fā)的幽門平滑肌張力(P<0.01),不抑制SP引發(fā)的幽門平滑肌振幅(P>0.05),,NK_1受體拮抗劑抑制SP引發(fā)的胃竇平滑肌張力(P<0.01)及振幅(P<0.01)。 結(jié)論:1.EFS增強(qiáng)幽門、胃竇平滑肌自發(fā)性收縮;2.BTX-A抑制幽門、胃竇平滑肌自發(fā)性收縮;3.BTX-A抑制EFS引發(fā)的幽門、胃竇平滑肌收縮;提示BTX-A可能通過(guò)抑制內(nèi)源性遞質(zhì)釋放或?qū)χ苯与妶?chǎng)刺激肌肉產(chǎn)生的收縮的抑制,從而抑制胃平滑肌的收縮。4.SP增強(qiáng)幽門、胃竇平滑肌收縮;5.BTX-A抑制SP引發(fā)的幽門、胃竇平滑肌收縮;6.NK_1受體拮抗劑抑制SP引發(fā)的幽門、胃竇平滑肌收縮,提示SP可能通過(guò)和平滑肌上受體結(jié)合發(fā)揮作用,使肌肉收縮增強(qiáng),而NK_1受體拮抗劑則可能通過(guò)拮抗SP與受體結(jié)合使肌肉收縮減弱。BTX-A可能通過(guò)抑制突觸囊泡與突觸前膜融合、胞吐,從而抑制遞質(zhì)釋放發(fā)揮作用,也可能對(duì)SP與突觸后膜上相應(yīng)受體結(jié)合具有一定的影響作用,導(dǎo)致對(duì)SP引發(fā)的幽門、胃竇離體平滑肌收縮的抑制。
