肝細粒棘球蚴動物模型的建立及骨橋蛋白在外囊壁中的表達

【摘要】: 目的:建立肝包蟲動物模型并觀察包蟲囊壁情況,研究骨橋蛋白在小鼠肝細粒棘球蚴囊壁周圍的分布特征及表達。 方法:選擇同窩同種小鼠180只,隨機分為免疫正常組及免疫抑制組,每組各90只,一組小鼠制作非特異性免疫抑制模型。將以上免疫正常及免疫抑制組按隨機的方法又分別分為1、2、3組,每組30只,分別采用門靜脈穿刺注射、開腹直視下肝臟穿刺注射及經皮肝臟穿刺注射細粒棘球蚴(Echinococcus granulosus,E.g)原頭節(jié)混懸液的方法,制備肝E. g動物模型,免疫正常組作HE染色觀察,免疫組化觀察小鼠肝細粒棘球蚴外囊壁中骨橋蛋白(Osteopontin ,OPN)的表達與分布。 結果:免疫正常組小鼠肝臟E. g感染率依據(jù)接種方法不同分別為93.1%、80.2%及59.3% ,免疫抑制組感染率分別為93.3%、79.3%及56.4%,兩組無明顯差異(P0.05)。HE切片染色顯示外囊與周圍組織具有明確界線,不存在漸變的過程, 83%肝細粒棘球蚴外囊壁外側有不同程度OPN表達,且分布與囊壁外側炎性細胞分布一致,在外囊層僅有少量OPN表達(P0.01)。 結論:門靜脈穿刺、開腹肝穿及經皮肝穿均可制備繼發(fā)性肝臟E. g動物模型,且免疫正常組與抑制組感染率、包囊直徑及包囊個數(shù)無明顯差別,其中門靜脈穿刺注射法較其他兩種方法接種感染率高,包囊個數(shù)多,為理想的肝E. g動物模型制備方法,同時在動物模型上證實了早期肝細粒棘球蚴外囊為肉芽腫反應結果, OPN主要分布在肝細粒棘球蚴壁囊外側炎性細胞中,并在外囊的形成過程中起重要作用。
[Abstract]:Aim: to establish an animal model of hepatic hydatid and to study the distribution and expression of osteopontin around the cyst wall of hepatic hydatid in mice. Methods: 180 homologous mice of the same nest were randomly divided into normal immune group and immunosuppressive group with 90 mice in each group. A non-specific immunosuppressive model was made in one group of mice. The above immune normal and immunosuppressive groups were randomly divided into 3 groups with 30 rats in each group. Each group was injected with portal vein puncture, liver puncture under open vision and percutaneous hepatic injection of (Echinococcus granulosus, (Echinococcus granulosus). E.G) the animal model of liver Eg was established by the suspension of proto-cephalic ganglion. The expression and distribution of osteopontin (Osteopontin, OPN) in the wall of hepatic hydatid of mice were observed by HE staining and immunohistochemistry. Results: according to the different inoculation methods, the liver Eg infection rate of the immunized normal mice was 93.2% and 59.3%, respectively. The infection rate of the immunosuppressive group was 93.3% and 56.4%, respectively, and that of the immunosuppressive group was 79.3% and 56.4%, respectively. There was no significant difference between the two groups (P0.05). HE staining showed that there was a clear boundary between the outer capsule and the surrounding tissues, and there was no gradual change. 83% of the patients had different degree of OPN expression in the outer wall of the epicystic wall of Echinococcus granulosus. The distribution was consistent with the distribution of inflammatory cells on the lateral wall of the capsule. There was only a small amount of OPN expression in the outer capsule (P0.01). Conclusion: portal vein puncture, open liver puncture and percutaneous liver puncture can make the animal model of secondary liver Eg, and there is no significant difference in infection rate, cyst diameter and number of cyst between immune normal group and inhibition group. Among them, the portal vein puncture injection method has higher infection rate and more cyst than the other two methods. It is an ideal method for the preparation of liver Eg animal model. At the same time, the results of granulomatous reaction of the early hepatic hydatid cyst were confirmed in the animal model. OPN mainly distributes in the inflammatory cells of the lateral wall of the hepatic hydatid wall and plays an important role in the formation of the outer capsule.
