TGFβ1介導新生大鼠心肌成纖維細胞Ⅰ型和Ⅳ型膠原合成的研究

【摘要】: 背景:心肌纖維化系心肌成纖維細胞增殖、細胞外基質合成及細胞凋亡等一系列復雜因素調控所產(chǎn)生的結果。在心肌纖維化的進程中,轉化生長因子-β_1(TGF-β_1)起著重要調控作用,它通過加速心肌成纖維細胞的分化、抑制心肌成纖維的凋亡、促進細胞外基質的產(chǎn)生而刺激纖維化反應。這些信號分子的作用受細胞的內環(huán)境的影響,且具有器官特異性,細胞與細胞之間以及細胞與基質問的相互作用也能促進或阻止該信號通路。迄今為止,以TGF-β_1干預心肌成纖維細胞的體外研究中,心肌成纖維細胞的生物學反應程度及其作用機制尚不清楚。因此,以TGF-β_1干預心肌成纖維細胞,通過檢測其下游通路的信號分子,,來探討其在心肌纖維化中的作用,對于改善心臟疾病的治療及降低疾病的纖維化所致的死亡具有重要意義。 目的:觀察TGF-β_1對原代培養(yǎng)的新生大鼠心肌成纖維細胞Ⅰ型與Ⅳ膠原表達的影響,探討TGF-β_1在心肌纖維化發(fā)生發(fā)展中的可能作用機制。 方法:取出生3-7天的SD大鼠心臟,利用酶消化法結合差速貼壁體外培養(yǎng)心肌成纖維細胞;分別用不同濃度的TGF-β_1(TGFβ1=0ng/ml、1ng/ml、10ng/ml)干預心肌成纖維細胞,分別于干預后5h、24h,采用RT-PCR及免疫細胞化學染色法檢測Ⅰ型膠原和Ⅳ型膠原的表達。 結果: 1、用胰酶與Ⅱ型膠原酶合用消化分離新生大鼠心肌成纖維細胞,可將心肌細胞和心肌成纖維細胞分離,心肌成纖維細胞抗波形蛋白(vimentin)染色陽性,抗α-橫紋肌肌動蛋白(α-sarcomeric actin)染色陰性。 2、用0ng/ml(對照組),1ng/ml和10ng/ml的TGF-β_1干預心肌成纖維細胞5h,RT-PCR檢測Ⅰ型膠原/β-actin光密度值分別為0.61±0.02,0.73±0.03和0.86±0.04,光密度值隨著TGFβ1濃度的增加而增加,各組比較,差異有顯著性(P<0.01);干預心肌成纖維細胞24h后,RT-PCR檢測Ⅰ型膠原/β-actin光密度值分別為0.65±0.03,0.91±0.02和0.98±0.02,光密度值隨著TGF-β_1濃度的增加而增加,各組比較,差異有顯著性(p<0.01)。 3、用0ng/ml(對照組),1ng/ml and 10ng/ml的TGF-β_1干預心肌成纖維細胞5h,RT-PCR檢測Ⅳ型膠原/β-actin光密度值分別為0.58±0.06,0.71±0.02,0.87±0.02,光密度值隨著TGF-β_1濃度的增加而增加,各組比較,差異有顯著性(p<0.01);干預心肌成纖維細胞24h后,RT-PCR檢測Ⅳ型膠原/β-actin光密度值分別為0.62±0.01,0.83±0.05,0.96±0.02,光密度值隨著TGF-β_1濃度的增加而增加,各組比較,差異有顯著性(p<0.01)。 4、用0ng/ml(對照組),1ng/ml and 10ng/ml的TGF-β_1干預心肌成纖維細胞5h,24h后,用免疫細胞化學染色法檢測心肌成纖維細胞Ⅰ型膠原和Ⅳ型膠原的表達情況,結果顯示,隨著濃度的增加,胞漿顏色由淺黃色變成深黃色。 結論: 1、成功分離和培養(yǎng)新生大鼠心肌成纖維細胞。 2、TGF-β_1可使心肌成纖維細胞Ⅰ型膠原和Ⅳ型膠原表達上調,呈時間和劑量依賴性。 3、TGF-β_1能促進心肌成纖維細胞Ⅰ型膠原和Ⅳ型膠原的合成,提示TGF-β_1在心肌纖維化中可能起了重要作用。
