人類精子培養(yǎng)液中能量成分對精子活動指標(biāo)的影響

【摘要】: 目的:探討精子培養(yǎng)液中能量成分的改變對人類精子活力及活率的影響 方法:精液取自自愿供精者,年齡25-40歲,禁欲3-7天,手淫法留取整份精液于無菌杯中,經(jīng)精液常規(guī)分析符合WHO規(guī)定的正常標(biāo)準(zhǔn){2},精漿、血清抗精子抗體均為陰性。以人類輸卵管液(Human Tube Fluid,HTF)為基本成分,對HTF配方中的能量成分進行改變作為實驗組,以HTF液作為對照組,觀察不同配方精子培養(yǎng)液對精子活力(A級精子、B級精子所占精子總數(shù)的百分率)、活率(A級精子、B級精子、C級精子所占精子總數(shù)的百分率)的影響。不同配方精子培養(yǎng)液分組如下:①實驗一:實驗組含果糖液(將果糖25mg替代HTF中的葡萄糖)及無糖液(不含糖的HTF)。②實驗二:在HTF中加入甘氨酸1.0mM、谷氨酰胺1.0mM、;撬0.1mM。③實驗三:在HTF中加入正辛酸0.329mM、正己酸0.38mM取代HTF中的丙酮酸。④實驗四:實驗組含ADP 2.5mM、含ATP 2.5mM。⑤實驗五:實驗組含肉毒堿2.5mM及含己酮可可堿2.5mM。每組取10份精液標(biāo)本,等分為2份或3份,用實驗組及對照組培養(yǎng)液進行處理。統(tǒng)計學(xué)處理采用SPSS10.0軟件包。 結(jié)果:①實驗一中于培養(yǎng)精子48小時的活力、活率及培養(yǎng)72小時的活力的P值0.05,果糖組優(yōu)于對照組,對照組優(yōu)于無糖組。其余時日的動力學(xué)指標(biāo)雖無統(tǒng)計學(xué)意義,但果糖組、對照組均優(yōu)于無糖組。②實驗二至實驗五中實驗組及對照組各項指標(biāo)P值0.05,差異無統(tǒng)計學(xué)意義。 結(jié)論:實驗表明,在HTF培養(yǎng)液中加入氨基酸、脂肪酸、ADP、ATP、肉毒堿及己酮可可堿均未明顯改善精子活力、活率。在實驗一中實驗組與對照組各項指標(biāo)無顯著差異,但隨著培養(yǎng)時間的延長,精子的活力、活率出現(xiàn)差異,果糖組優(yōu)于對照組,對照組優(yōu)于無糖組。提示六碳糖是精子的重要供能物質(zhì),以果糖代替葡萄糖可顯著改善精子的活動能力。
[Abstract]:Objective: to investigate the effect of the changes of energy components in sperm culture medium on the motility and motility of human spermatozoa methods: semen was obtained from volunteers aged 25-40 years, abstinence 3-7 days. The whole semen was collected by masturbation in the aseptic cup. The sperm routine analysis met the normal standard {2} stipulated by WHO. The seminal plasma and serum anti-sperm antibody were all negative. Human oviduct fluid (Human Tube Fluid,HTF) was used as the basic component, the energy composition of HTF formula was changed as the experimental group, and the HTF solution was used as the control group. The sperm motility (A grade spermatozoa) was observed in different formulations of spermatozoa culture medium. The effect of the percentage of B grade spermatozoa on the total number of spermatozoa and the percentage of A grade spermatozoa, B grade sperm and C grade spermatozoa in the total number of spermatozoa. Different formulations of spermatozoa culture fluid were grouped as follows: experiment 1: fructose solution (fructose 25mg was substituted for glucose in HTF) and sugar-free solution (HTF). 2 without sugar); experiment 2: adding glycine 1.0 mm, glutamine 1.0 mm in HTF, glutamine 1.0 mm, Glutamine 1.0 mm. Taurine 0.1mM.3 experiment 3: the addition of n-octanoic acid 0.329mm in HTF and the substitution of pyruvate in HTF by n-caproic acid 0.38mM. 4 experiment 4: the experimental group contains ADP 2.5mM. Experiment 5 with ATP 2.5mM.5: the experimental group contained carnitine 2.5mM and pentoxifylline 2.5 mm. In each group, 10 semen samples were collected and divided into 2 or 3 semen samples, which were treated with the culture medium of the experimental group and the control group. SPSS10.0 software package was used for statistical processing. Results: 1 in experiment 1, the motility and viability of spermatozoa cultured for 48 hours and the activity of 72 hours were 0.05. The fructose group was superior to the control group, and the control group was superior to the sugar-free group. Although there was no statistical significance in the other days, the fructose group and the control group were better than the non-sugar group. 2 from experiment 2 to experiment 5, there was no significant difference between the experimental group and the control group (P 0.05). Conclusion: the addition of amino acids, fatty acids, ADP,ATP, carnitine and pentoxifylline into HTF medium did not significantly improve sperm motility and motility. In experiment 1, there was no significant difference between experimental group and control group, but with the extension of culture time, sperm motility and viability were different. Fructose group was superior to control group, and control group was superior to sugar-free group. It is suggested that hexacarbonose is an important energy supplying substance in spermatozoa. Fructose instead of glucose can significantly improve the motility of spermatozoa.
