母胎界面L-選擇素和其配體表達及對胚胎著床的影響

【摘要】:在生殖、免疫、炎癥的一些關鍵性過程中,分子水平上存在相似的特征。黏附分子L-選擇素與其配體的結合介導了免疫系統(tǒng)的淋巴細胞歸巢,及炎癥過程白細胞與內皮細胞的黏附,并發(fā)揮著信號分子作用。近年研究者發(fā)現(xiàn),參與免疫和炎癥過程的重要細胞黏附分子L-選擇素及其配體也存在于哺乳動物著床期母胎界面上。Genbacev等發(fā)現(xiàn)L-選擇素與其配體的結合可能構成人類胚胎著床的起始步驟。著床窗口期,胚胎表面的L-選擇素和母體子[諛-選擇素的寡糖配體似乎都呈現(xiàn)上調趨勢。L-選擇素配體至少包括四種分子:CD34、粘膜區(qū)素細胞粘附分子-1(MAdCAM-1)、依賴糖基化的細胞粘附分子-1(GlyCAM-1)、Sgp200;由于最早發(fā)現(xiàn)于小鼠淋巴結高內皮靜脈,故將它們統(tǒng)稱為周圍淋巴結區(qū)素(PNAd)。L-選擇素識別硫酸化形式的配體,N-乙酰葡萄糖胺-6-轉磺酶(GlcNAc6ST)在功能型L-選擇素配體生成過程中發(fā)揮重要作用。 盡管以往的研究發(fā)現(xiàn)了人類母胎界面上存在L-選擇素和其配體,但有關早期胚胎發(fā)育過程中L-選擇素的表達規(guī)律、周期性變化的子[諛ど螸-選擇素配體表達的時間、空間模式、妊娠建立后它們又將如何變化、它們的表達水平是否與妊娠成功率有關,這一系列問題目前尚未見報道。盡管體外研究表明這對配體一受體確實能夠介導細胞之間的黏附,但尚無體內實驗證明它們在胚胎著床中的作用。為了深入探討L-選擇素系統(tǒng)在胚胎著床中的功能和影響,我們檢測人類母胎界面L-選擇素及其配體表達的時間規(guī)律、位置特征,分析分泌期子[諛-選擇素配體(PNAd)各組成分子以及GlcNAc6ST mRNA水平與體外
[Abstract]:There are similar characteristics at the molecular level in some key processes of reproduction, immunity, and inflammation. The binding of the adhesion molecule L- selectin to its ligand mediates the homing of lymphocytes in the immune system and the adhesion of leukocytes to endothelial cells during inflammation and acts as a signaling molecule. In recent years, researchers have found that L- selectin and its ligands, which are important cell adhesion molecules involved in immune and inflammatory processes, also exist at mammalian maternal and fetal interfaces. Genbacev et al found that the binding of L- selectin to its ligands may constitute the initial step of human embryo implantation. L- selectin and mother on embryo surface during implantation window period [? Flat-selectin oligosaccharide ligands appear to be on an upward trend. Lselectin ligands contain at least four molecules: CD34, mucosal adhesion molecule-1 (MAdCAM-1), glycosylated cell-adhesion molecule-1 (GlyCAM-1), and glycosylated oligosaccharide ligands. Although in vitro studies have shown that these ligands and receptors can indeed mediate cell adhesion, no in vivo studies have demonstrated their role in embryo implantation. In order to study the function and influence of L- selectin system in embryo implantation, we detected the expression of L- selectin and its ligand in human maternal and fetal interface, and analyzed the secretory phase of L- selectin and its ligand.
