抑制泛素-蛋白酶體途徑對(duì)晶狀體上皮細(xì)胞自噬的誘導(dǎo)作用

【摘要】:目的在晶狀體上皮細(xì)胞中通過(guò)蛋白酶抑制劑MG132抑制泛素-蛋白酶體途徑活性,探索其對(duì)自噬作用的影響及相關(guān)機(jī)制。方法使用蛋白酶抑制劑MG132作用于晶狀體上皮細(xì)胞,阻斷泛素-蛋白酶體途徑的功能,通過(guò)蛋白免疫印跡法檢測(cè)泛素化蛋白質(zhì)水平評(píng)價(jià)泛素-蛋白酶體系統(tǒng)活性。蛋白免疫印跡法觀察自噬作用活性標(biāo)志蛋白LC3-Ⅱ的表達(dá)情況,共聚焦免疫熒光顯微鏡下觀察LC3、p62的表達(dá)與分布。蛋白免疫印跡法檢測(cè)p62表達(dá)的變化。結(jié)果在MG132作用后,晶狀體上皮細(xì)胞中泛素化蛋白聚集物明顯增加。隨著抑制時(shí)間的延長(zhǎng),LC3-Ⅱ的表達(dá)量也隨之增加,自噬作用活性增強(qiáng),p62的表達(dá)也明顯增加。結(jié)論在晶狀體上皮細(xì)胞中,當(dāng)泛素-蛋白酶體系統(tǒng)活性受抑制時(shí)自噬作用活性增強(qiáng)。
