大鼠異體腎移植動物模型的建立及術后外周血細胞因子的動態(tài)監(jiān)測

【摘要】:目的:建立新疆地區(qū)第一個大鼠異體腎移植動物模型,為腎移植基礎研究提供良好的技術平臺。在成功建模的基礎上,觀察異體腎移植急性排斥大鼠術后外周血中IL-2、IL-4和IFN-γ的含量變化并探討其意義。方法:采用遠交系Wistar和SD大鼠,血管吻合采用供受體腹主動脈和下腔靜脈端-側連續(xù)縫合,尿路重建采用供受體輸尿管端-端吻合,建立大鼠異體腎移植模型,并觀察和比較Wistar-Wistar與SD-Wistar兩組大鼠間異體腎移植后的存活情況,術后并發(fā)癥及腎臟病理損害。把制備的大鼠模型分為四組,即正常對照組、假手術組、同系移植組和異系移植組,分批采集假手術組、正常對照組和同系及異系移植組大鼠在術后3d、5d、7d的外周血,監(jiān)測鼠外周血中IL-2、IL-4、IFN-γ、Cr和BUN的動態(tài)改變,同時行移植腎病理檢查。結果:大鼠異體腎移植手術時間平均為108±15min,手術成功率為83.33%(25/30)。對術后急性排斥大鼠體內的細胞因子的監(jiān)測表明,IL-2、IFN-γ在急排早期明顯升高,隨后緩慢下降,IL-4雖有升高但比較正常對照組無統計學差異(P0.05)。發(fā)生急性排斥反應時,IL-2、IFN-γ的升高和移植腎病理損害程度無相關關系。Cr、BUN的變化和移植腎病理損害程度呈正相關(P0.05)。結論:本組大鼠異體腎移植動物模型成功率較高,并發(fā)癥發(fā)生率較低,能滿足實驗研究的需要。急性排斥反應發(fā)生時,大鼠外周血中IL-2、IFN-γ的升高有助于早期診斷急性排斥反應,其升高水平與移植腎急性排斥病變程度無相關關系。在本研究中尚無證據支持IL-4的含量變化可以作為早期診斷急性排斥反應的依據。
[Abstract]:Aim: to establish the first animal model of allogeneic kidney transplantation in Xinjiang, and to provide a good technical platform for the basic research of renal transplantation. On the basis of successful modeling, the changes of IL-2,IL-4 and IFN- 緯 levels in peripheral blood of acute rejection rats after allograft transplantation were observed and their significance was discussed. Methods: a rat model of allograft kidney transplantation was established by end-to-side anastomosis of donor abdominal aorta and inferior vena cava and end-to-end ureteral anastomosis for urinary tract reconstruction in distal Wistar and SD rats. The survival, postoperative complications and renal pathological damage were observed and compared between Wistar-Wistar group and SD-Wistar group. The rat models were divided into four groups: normal control group, sham operation group, homologous transplantation group and allogeneic transplantation group, batch collection of sham operation group, normal control group, homologous group and allogeneic transplantation group. The dynamic changes of IL-2,IL-4,IFN- 緯, Cr and BUN in peripheral blood of rats were monitored. Results: the average time of allograft kidney transplantation was 108 鹵15 min, and the success rate was 83.33% (25 / 30). The monitoring of cytokines in acute rejection rats after operation showed that IL-2,IFN- 緯 increased significantly at the early stage of acute rejection, and then decreased slowly. Although IL-4 was increased, there was no statistical difference compared with the normal control group (P0.05). When acute rejection occurred, there was no correlation between the increase of IL-2,IFN- 緯 and the degree of pathological damage of transplanted nephropathy. The change of Cr,BUN was positively correlated with the degree of pathological damage of transplanted nephropathy (P0.05). Conclusion: the success rate of allogeneic kidney transplantation in this group is high, and the incidence of complications is low, which can meet the need of experimental study. When acute rejection occurs, the increase of IL-2,IFN- 緯 in peripheral blood of rats is helpful to early diagnosis of acute rejection, and there is no correlation between the elevated level and the severity of acute rejection of transplanted kidney. There is no evidence in this study that changes in IL-4 levels can be used as a basis for early diagnosis of acute rejection.
