賀斯在體外對健康人免疫系統(tǒng)細(xì)胞因子的影響
[Abstract]:Objective to investigate the effects of different concentrations of hydroxyethyl starch plasma substitutes (HES 200 / 0.5) on the levels of interleukin-2siloxacin 1012 (IL-2m10N12) and interferon- 緯 (IFN- 緯) in normal human immune system in order to determine whether they inhibit the immune system of normal human beings. In order to reduce the recurrence rate, postoperative infection rate and mortality of malignant tumor caused by blood transfusion, we can provide experimental basis for replacing blood transfusion in perioperative period. Methods Mononuclear cells were isolated from 12 normal volunteers under strict aseptic conditions by extracting 20 ml of venous blood and using heparin as anticoagulant. After counting, the cells were divided into 5 groups, 10% plasma control group, 10% Horse test group and 40% Horse test group, then each group was divided into negative control group, lipopolysaccharide (LPS) group and phytohemagglutinin (PHA) group. After the mononuclear cells were cultured in different culture medium for 48 hours, the supernatants were extracted by centrifugation. The concentrations of cytokines interleukin-210-10 (IL-12) and interferon- 緯 (IFN- 緯) in the supernatants were detected by enzyme-linked immunosorbent assay (ELISA). Results the concentrations of four cytokines were slightly different between the three concentrations of Horse test group and 1640 medium and plasma control group in three environments (negative control, plus LPS, plus PHA), although the concentrations of four cytokines were slightly different. In IL-2 and IFN- 緯, there was no significant difference between the negative control group and the LPS group (p0.05), but there was a significant difference between the two groups (p0.05), and there was a significant difference between the IL-10 IL-12 group, the negative control group, the LPS group and the PHA group (p0.05). Conclusion there was no significant difference in cytokine concentrations between the three concentrations of Hos cultured mononuclear cells in vitro and plasma and negative medium groups after 48 hours, suggesting that the three concentrations of Hos did not affect the survival of immune cells in vitro.
