肌成纖維細(xì)胞在大鼠排卵機(jī)制中的作用

【摘要】:目的: 1.確認(rèn)在大鼠卵巢卵泡膜中有肌成纖維細(xì)胞存在 2.探討肌成纖維細(xì)胞與卵巢排卵的關(guān)系 方法: 1.采用免疫組織化學(xué)方法檢測(cè)α-SMA、desmin、vimentin、ET-AR和ET-BR在動(dòng)情期組(n=20)與非動(dòng)情期組(n=10)大鼠卵巢各級(jí)卵泡(竇前卵泡、小竇狀卵泡和大竇狀卵泡)卵泡膜細(xì)胞中的表達(dá)情況,并進(jìn)行圖像分析。 2.采用透射電鏡技術(shù)觀察動(dòng)情期大鼠竇狀卵泡卵泡膜細(xì)胞的超微結(jié)構(gòu)。 結(jié)果: 1.免疫組織化學(xué)結(jié)果: 1.1.α-SMA表達(dá)于動(dòng)情期和非動(dòng)情期各級(jí)卵泡的卵泡膜細(xì)胞中,其表達(dá)強(qiáng)度在組間同級(jí)卵泡間比較有顯著性差異(P<0.05),組內(nèi)各級(jí)卵泡間比較也有顯著性差異(P<0.05);在動(dòng)情期組其表達(dá)強(qiáng)度與卵泡直徑大小成正相關(guān)(r=0.631),而非動(dòng)情期組兩者卻幾乎無(wú)相關(guān)性(r=-0.087)。 1.2.Desmin在動(dòng)情期組各級(jí)卵泡的卵泡膜細(xì)胞中表達(dá),其表達(dá)強(qiáng)度在該組各級(jí)卵泡間比較有顯著性差異(P<0.05):在非動(dòng)情期組各級(jí)卵泡的卵泡膜細(xì)胞中無(wú)表達(dá)。 1.3.Vimentin表達(dá)于動(dòng)情期和非動(dòng)情期各級(jí)卵泡的卵泡膜細(xì)胞中,其表達(dá)強(qiáng)度在組間同級(jí)卵泡間比較有顯著性差異(P<0.05)。動(dòng)情期組各級(jí)卵泡間比較有顯著性差異(P<0.05)而且vimentin表達(dá)強(qiáng)度與卵泡直徑大小呈正相關(guān)(r=0.536);非動(dòng)情期組竇前卵泡和小竇狀卵泡間比較無(wú)明顯差異(P>0.05),其他級(jí)別卵泡間比較有顯著性差異(P<0.05),而且vimentin表達(dá)強(qiáng)度與卵泡
[Abstract]:Objective: 1. To confirm the presence of myofibroblasts in the follicular membrane of rat ovary 2. To investigate the relationship between myofibroblasts and ovulation. 1.Immunohistochemical method was used to detect the ovarian follicles (preantral follicles) in the estrus group (nong20) and the non-estrous group (nm10). The expression of follicular membrane cells in antral follicles and large antral follicles, The ultrastructure of antral follicular membrane cells in estrus was observed by transmission electron microscopy. Results: 1. Immunohistochemical results: 1.1. 偽 -SMA was expressed in follicular membrane cells of all levels of follicles in estrus and non-estrous stage. There was a significant difference in the expression intensity between the same grade follicles (P < 0.05) and among the follicles of different levels in the group (P < 0.05). There was a positive correlation between the expression intensity and follicle diameter in estrus group (r 0. 631), but almost no correlation between the expression intensity and follicle diameter in non estrus group (r 0. 087). 1.2.Desmin was active in oestrus. In the oestrous stage group, the follicular membrane cells of all levels of follicles were expressed, There was significant difference in the expression intensity among the follicles (P < 0.05), and there was no expression in the theca cells of the follicles in the non-estrous period group. 1.3.Vimentin was expressed in follicular membrane cells of all levels of follicles in estrus and non-estrous stage.
