滅活的雙歧桿菌抗腸源性感染的實(shí)驗(yàn)研究

【摘要】: 雙歧桿菌作為人和動物腸道中最重要的生理性細(xì)菌之一,對機(jī)體無毒、無害、并且具有廣泛的生態(tài)效應(yīng)與生理作用。雙歧桿菌參與了宿主的消化、營養(yǎng)、代謝、吸收、免疫及抗感染過程,被認(rèn)為是微生態(tài)學(xué)研究的核心或重心�,F(xiàn)有研究表明,雙歧桿菌制劑可恢復(fù)腸道內(nèi)菌群失調(diào)的生態(tài)平衡,保護(hù)并修復(fù)腸道黏膜,促進(jìn)腸道益生菌增殖,因而在防治腸源性感染中起到了良好的治療效果。目前國內(nèi)外研制開發(fā)的雙歧桿菌制劑采用的都是活菌,其有效期不過一年左右,這樣就限制了此類產(chǎn)品的應(yīng)用范圍與條件。如果我們將滅活的雙歧桿菌制成微生態(tài)調(diào)節(jié)劑,進(jìn)行工業(yè)化生產(chǎn),通過提高產(chǎn)品質(zhì)量來增強(qiáng)其競爭力,,就可以打破國外此類產(chǎn)品對國內(nèi)市場的壟斷,因此本研究對推動微生態(tài)學(xué)基礎(chǔ)理論研究有著重要意義。 本實(shí)驗(yàn)將雙歧桿菌接種至BS肉湯培養(yǎng)基中,37℃厭氧培養(yǎng)48h。所得菌懸液分為活菌組和死菌組。將活、死兩組菌懸液用NS洗3次,離心棄上清,用SCS重懸活、死雙歧桿菌沉淀,使菌懸液濃度均達(dá)到1×10~(11) CFU/mL。將昆明種小鼠隨機(jī)分為活菌組、死菌組、自然恢復(fù)組、正常對照組4組,每組10只。除正常對照組外,其余3組小鼠分別造成抗生素致菌群失調(diào)、環(huán)磷酰胺致免疫功能低下及燒傷動物模型。從第4d開始,活菌組采用雙歧桿菌活菌懸液灌胃;死菌組采用雙歧桿菌滅活菌懸液灌胃;自然恢復(fù)組采用NS灌胃,連續(xù)3d。4組小鼠分別摘眼球采血處死,利用日本OLYPUS-AU400全自動生化儀對臨床血清學(xué)指標(biāo)進(jìn)行測定。 實(shí)驗(yàn)結(jié)果如下: 1.滅活的雙歧桿菌治療小鼠菌群失調(diào)致腸源性感染 采用腹腔注射青霉素造成抗生素菌群失調(diào)小鼠動物模型,分別以雙歧桿菌滅活菌液、活菌液對小鼠進(jìn)行灌胃治療后,對血清總蛋白(TP)、血清白蛋白(ALB)、血清球蛋白(GLO)、y-谷氨酰氨轉(zhuǎn)肽酶(GGT)、血清尿素氮(BUN)、乳酸脫氫酶(LDH)、血肌酐(CRE)以及組織勻漿內(nèi)的示蹤菌進(jìn)行測定。結(jié)果顯示,死菌組與自然恢復(fù)組比較,TP、GLO明顯升高,GGT、BUN則顯著降低。死菌組與活菌組、正常對照組比較,LDH明顯降低,死菌組與自然恢復(fù)組比較,腎、肝、肺組織勻漿內(nèi)腸桿菌數(shù)量均明顯下降。 2.滅活的雙歧桿菌治療免疫功能低下致小鼠腸源性感染 采用腹腔注射環(huán)磷酰胺造成免疫功能低下小鼠動物模型,分別以雙歧桿菌滅活菌液、活菌液對小鼠進(jìn)行灌胃治療后,檢測臨床血清學(xué)指標(biāo)與組織勻漿內(nèi)示蹤菌。結(jié)果顯示,活菌組、死菌組同自然恢復(fù)組相比,臨床血清學(xué)指標(biāo)水平與組織勻漿內(nèi)示蹤菌數(shù)量恢復(fù)至正常對照組水平。 3.滅活的雙歧桿菌治療燒傷致小鼠腸源性感染 制作Ⅲ度燙傷(燒傷)小鼠動物模型,分別以雙歧桿菌滅活菌液、活菌液對小鼠進(jìn)行灌飼后,檢測組織勻漿內(nèi)示蹤菌數(shù)量與血清蛋白水平。結(jié)果顯示,活菌組、死菌組同自然恢復(fù)組相比,組織勻漿內(nèi)示蹤菌數(shù)量與血清蛋白水平恢復(fù)至正常對照組水平。 結(jié)論: 1.雙歧桿菌滅活菌懸液具有與活菌懸液相同的抗菌群失調(diào)致小鼠腸源性感染作用。 2.雙歧桿菌滅活菌液具有與活菌液相同的抗免疫功能低下致小鼠腸源性感染作用。 3.雙歧桿菌滅活菌液具有與活菌液相同的治療小鼠燒傷感染作用。
[Abstract]:Bifidobacterium as one of the most important physiological bacteria in human and animal intestinal tract, harmless to the body, non-toxic, and has ecological effects and extensive physiological functions. Bifidobacteria participate in host digestion, nutrition, metabolism, absorption, immunity and anti infection, is considered to be the core of micro ecology research or existing focus. Studies show that bifidobacteria can restore intestinal flora ecological balance, protection and repair of the intestinal mucosa, promote the proliferation of probiotics, which played a good therapeutic effect in the prevention of enterogenic infection. At home and abroad developed by Bifidobacterium are viable, the period of validity but a year or so, thus limiting the application scope and conditions of such products. If we made the inactivated Bifidobacterium micro ecological regulator, industrial production, to increase by improving the quality of products Strong competitiveness can break the monopoly of foreign products to the domestic market. Therefore, this study is of great significance to the research of the basic theory of microecology.
The bifidobacteria were inoculated in BS broth, 37 C anaerobic bacteria suspension culture of 48h. income divided into viable group and death group. The live bacteria, dead bacteria suspension with two were washed 3 times with NS, centrifuged supernatant, with SCS suspending live Bifidobacterium bacteria to death precipitation. The suspension concentration reached 1 * 10~ (11) CFU / mL. Kunming mice were randomly divided into group of live bacteria, dead bacteria group, natural recovery group, normal control group 4 groups, 10 rats in each group. Except the normal control group, the remaining 3 groups of mice were caused by antibiotic induced dysbacteriosis, cyclophosphamide induced immune hypofunction and burn animal model. From the beginning of 4D, bacteria group with Bifidobacterium suspension gavage; dead bacteria group using Bifidobacterium inactivated bacteria suspension gavage; natural recovery group gavaged with NS, continuous 3d.4 mice were eyeball blood were killed, the clinical blood indexes using OLYPUS-AU400 automatic biochemical analyzer Determination.
The experimental results are as follows:
1. inactivated bifidobacteria in the treatment of intestinal dysbacteria induced intestinal infection in mice
By intraperitoneal injection of penicillin antibiotics dysbacteriosis caused by the animal model of mice were Bifidobacterium inactivated and live bacteria into the stomach of the mice after treatment, the serum total protein (TP), serum albumin (ALB), serum globulin (GLO), y- glutamine transpeptidase (GGT), serum urea nitrogen (BUN), lactate dehydrogenase (LDH), serum creatinine (CRE) and tissue homogenate in tracer bacteria were determined. The results show that the death group, bacteria group and natural recovery of TP, GGT, GLO increased significantly, BUN decreased significantly. The dead bacteria group and normal bacteria group. Compared to the control group, LDH decreased significantly, the death group, bacteria group and natural recovery of kidney, liver and lung homogenate in enterobacteria decreased significantly.
2. inactivated bifidobacteria in the treatment of intestinal infection in mice induced by immunodeficiency
By intraperitoneal injection of cyclophosphamide caused by low immune function mice animal model, respectively Bifidobacterium inactivated and live bacteria into the stomach of the mice after treatment, detection of clinical serum markers and tracer bacteria within tissue homogenates. The results showed that the living bacteria group, dead bacteria group and natural recovery group compared to the level of clinical and serological indicators the number of bacteria in the tissue homogenate tracer recovered to the normal level of the control group.
3. inactivated Bifidobacterium for the treatment of intestinal infection in mice caused by burn
Production of scald (burn) animal model of mice were Bifidobacterium inactivated and live bacteria liquid on mice after intragastric administration, the number of bacteria detection tracer homogenate and serum protein level. The results showed that the living bacteria group, dead bacteria group and natural recovery group compared to tissue homogenate of cytoplasm of tracer bacteria the number and serum protein levels returned to the normal level of the control group.
Conclusion:
1. the suspension of Bifidobacterium inactivated bacteria has the effect of intestinal infection in mice caused by the same antibacterial group as the living bacteria suspension.
2. Bacillus Bifidobacterium inactivated bacteria has the same anti immune function as the living bacteria, which induces intestinal infection in mice.
3. Bifidobacterium inactivated bacteria have the same effect as living bacteria solution in the treatment of burn infection in mice.
