【摘要】：一、背景： 患有酒精綜合癥(fetal alcohol syndrome)的兒童是在胚胎期和(或)哺乳期由母親過量飲酒行為引起，有明顯小于健康兒童的大腦及其他畸形。為研究在胎兒期大腦皮層神經(jīng)細胞在增殖、分化過程中受酒精損傷的原理，用培養(yǎng)的神經(jīng)前體細胞(neural progenitor cells)建立實驗?zāi)Ｐ�，以研究酒精對生長因子刺激引起神經(jīng)前體細胞數(shù)量擴增的影響。該模型的建立可幫助了解酒精綜合癥對神經(jīng)系統(tǒng)(central nervous system，CNS)損傷的機制，為保護暴露于酒精危害下胎兒的神經(jīng)系統(tǒng)發(fā)育提供預(yù)防及治療的科學(xué)依據(jù)。 神經(jīng)干細胞(neural stem cells，NSC)培養(yǎng)技術(shù)的發(fā)展，為體外研究神經(jīng)干細胞增殖、分化特性及神經(jīng)干細胞損傷保護提供了重要的實驗手段。國外已有報道貼壁培養(yǎng)神經(jīng)干細胞的方法，與傳統(tǒng)的神經(jīng)干細胞懸浮培養(yǎng)相比，更適于研究酒精對神經(jīng)前體細胞增殖、分化的影響，如免疫熒光染色、細胞計數(shù)等。 另外，，在神經(jīng)干細胞克隆球超微結(jié)構(gòu)觀察中發(fā)現(xiàn)，在克隆球中相鄰的兩個細胞膜之間有成片的連接，有2-4nm狹窄的縫隙，為間隙連接(gap junction)。構(gòu)成這種細胞間通道的蛋白質(zhì)分子被稱為連接蛋白(connexin)或者連接子(connexon)。連接蛋白基因是一個多基因家族，與神經(jīng)干細胞的增殖分化有關(guān)。確定神經(jīng)干細胞克隆球細胞之間連接蛋白的類型，可幫助了解神經(jīng)干細胞在體外培養(yǎng)以及中樞神經(jīng)系統(tǒng)(central nervous system，CNS)發(fā)育過程中間隙連接的演變過程及其規(guī)律。 國外新近報道，神經(jīng)前體細胞株在分化過程中能夠分泌某種抑制神經(jīng)膠質(zhì)瘤生長的因子。本試驗對神經(jīng)干細胞分化過程中產(chǎn)生的衍生物對神經(jīng)膠質(zhì)瘤細胞的
[Abstract]:I. background:. Children with alcohol syndrome, fetal alcohol syndrome, are caused by excessive drinking by their mothers during embryonic and / or lactating periods, with brain and other deformities that are significantly smaller than those of healthy children. On the principle of alcohol injury during differentiation, an experimental model was established by using cultured neural progenitor cells. In order to study the effect of alcohol on the proliferation of neural precursor cells induced by growth factor stimulation, the establishment of the model may help to understand the mechanism of alcohol syndrome damage to central nervous system of the nervous system. To provide the scientific basis of prevention and treatment for protecting the nervous system development of fetus exposed to alcohol. The development of neural stem cell culture provides an important experimental means for the study of neural stem cell proliferation, differentiation characteristics and injury protection of neural stem cells in vitro. Compared with the traditional suspension culture of neural stem cells, it is more suitable to study the effects of alcohol on the proliferation and differentiation of neural precursor cells, such as immunofluorescence staining, cell count and so on.
