體外誘導(dǎo)大鼠骨髓干細(xì)胞分化為肝樣細(xì)胞的實(shí)驗(yàn)研究

【摘要】:目的:探討大鼠骨髓干細(xì)胞在肝細(xì)胞生長因子(HGF)誘導(dǎo)下分化為肝樣細(xì)胞的可能性及分化特性。方法:HGF體外誘導(dǎo)大鼠骨髓干細(xì)胞向肝樣細(xì)胞分化,誘導(dǎo)第O、7、14、21天,觀察細(xì)胞形態(tài)特征;免疫細(xì)胞化學(xué)法檢測甲胎蛋白(AFP)、細(xì)胞角質(zhì)蛋白18(CKl8)的表達(dá);PAS檢測細(xì)胞糖原;逆轉(zhuǎn)錄聚合酶鏈反應(yīng)(RT-PCR)法檢測AFP mRNA、ALB mRNA水平的表達(dá)。結(jié)果:①新鮮分離的骨髓干細(xì)胞AFP mRNA呈弱陽性,ALB mRNA陰性。在非誘導(dǎo)組:AFPmRNA、ALB mRNA、AFP、CK18、糖原在各時(shí)間點(diǎn)均呈陰性。HGF誘導(dǎo)組:7天時(shí)AFP mRNA、AFP表達(dá)明顯增加,但此后表達(dá)逐漸減少ALB mRNA、CK18、糖原隨誘導(dǎo)時(shí)間表達(dá)逐漸增強(qiáng)。②HE染色較深的小圓形細(xì)胞在非誘導(dǎo)條件下其直徑未發(fā)生明顯變化;誘導(dǎo)組HE染色較深的小圓形細(xì)胞與AFP、CK18、糖原染色陽性的細(xì)胞形態(tài)大小一致,隨誘導(dǎo)時(shí)間延長其直徑增大。結(jié)論:①大鼠骨髓干細(xì)胞在HGF誘導(dǎo)下分化為肝樣細(xì)胞,肝細(xì)胞標(biāo)志物表達(dá)與肝細(xì)胞發(fā)育過程吻合。②骨髓源性肝干細(xì)胞的形態(tài)學(xué)特征為集落中HE染色較深的小圓形細(xì)胞。
[Abstract]:Objective: to investigate the possibility and characteristics of rat bone marrow stem cells differentiating into hepatoid cells induced by hepatocyte growth factor (HGF). Methods: rat bone marrow stem cells were induced to differentiate into hepatocyte-like cells in vitro, and OF71421 days were induced. The morphological characteristics of the cells were observed, the expression of AFP and CKl8 were detected by immunocytochemistry and the glycogen was detected by pas. Reverse transcriptase polymerase chain reaction reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of AFP mRNA-ALB mRNA in bone marrow stem cells isolated from fresh bone marrow stem cells. Results AFP mRNA of bone marrow stem cells isolated from freshly isolated bone marrow stem cells was weakly positive for ALB mRNA negative. In the non-induced group, AFP mRNA-ALB mRNA-AFPmRNA-AFPmRNA-AFPCK18, glycogen was negative at all time points .HGF-induced AFP mRNA was negative for 7 days. The expression of AFP mRNA-AFP was significantly increased. After that, the expression of ALB mRNA-CK18 was gradually decreased, and the diameter of the small circular cells stained deeply with the induction time was not changed significantly. In the induction group, the size of the small round cells stained deeply by HE was the same as that of AFP-CK-18, and the size of the cells with positive glycogen staining was the same, and the diameter of the cells increased with the prolongation of the induction time. Conclusion: the bone marrow stem cells of rats with HGF can differentiate into hepatoid cells. The expression of hepatocyte markers coincides with the development of hepatocytes. 2 the morphological characteristics of bone marrow-derived hepatic stem cells are small round cells stained deeply by HE in the colony.
