動(dòng)物腦內(nèi)酸性肽的鑒別和大鼠腦內(nèi)含量的分析

【摘要】:目的:鑒定酸性肽(acidic peptide,AP)在動(dòng)物腦中是否普遍存在,測(cè)定大鼠腦內(nèi)酸性肽和谷氨酸(glutamic acid,Glu)的含量,并探討外源性酸性肽對(duì)大鼠內(nèi)源性酸性肽及谷氨酸含量的影響。 方法:用紙電泳法(300V,1h)分離定位marker,此marker包括谷氨酸、酸性肽和谷胱甘肽三種標(biāo)準(zhǔn)小肽或氨基酸,然后用此marker鑒別牛、羊、雞、鴨、兔、大鼠等多種動(dòng)物腦中AP的存在。用標(biāo)準(zhǔn)谷氨酸、酸性肽和谷胱甘肽配成濃度分別為0.5mg/ml、1.0mg/ml、2.0mg/ml、3.0mg/ml、4.0mg/ml的混合液,每一濃度各取4μl點(diǎn)樣,然后進(jìn)行電泳分離,在烘箱中烘10min,剪下斑點(diǎn),洗脫液洗脫,分光光度法測(cè)定不同濃度的AP和Glu的吸光度值(OD)。重復(fù)10次實(shí)驗(yàn)。以平均OD值為縱坐標(biāo),濃度或含量為橫坐標(biāo),繪制AP和Glu的標(biāo)準(zhǔn)曲線(xiàn)。將大鼠隨機(jī)分為6組(每組10只),其中1組為對(duì)照組,其余5組按100mg/kg酸性肽灌胃,分別在灌胃后1h、
[Abstract]:Aim: to determine the prevalence of acidic peptide (APD) in animal brain, to determine the contents of acidic peptide and glutamic acid acid acid (glutamic acid acid) in rat brain, and to investigate the effect of exogenous acidic peptide on the content of endogenous acid peptide and glutamate in rats. Methods: Marker was isolated by paper electrophoresis for 1 h. The marker consisted of glutamic acid, acid peptide and glutathione. The marker was used to identify cattle, sheep, chicken, duck and rabbit. The presence of AP in the brains of rats and other animals.
