【摘要】：胰蛋白酶是一種動物來源的蛋白水解酶,具有高度的專一性,只水解L-型精氨酸和L-型賴氨酸的羧基組成的肽鍵。胰蛋白酶是所有胰臟蛋白酶原的共同激活劑(包括其自身的前體胰蛋白酶原)。作為一種重要的消化酶,胰蛋白酶在醫(yī)藥、食品、工業(yè)及有機合成中都有著廣泛的應(yīng)用,人們也對其進行了深入的研究。現(xiàn)在的熱點主要集中于天然胰蛋白酶抑制劑。 生物堿是一類含氮雜環(huán)的堿性天然有機物,廣泛存在于植物體內(nèi),大多數(shù)具有重要的醫(yī)療價值。本文基于生物堿的藥用價值,借助近年來發(fā)展迅速的光譜分析方法探討了三種生物堿小分子與胰蛋白酶的相互作用。 本文選擇的三種生物堿分別為:葛根素、秋水仙堿和喜樹堿。葛根素(puerarin)是由豆科植物野葛(pueraria lobata)甘藹藤(puerarin thomsonii)根中提取的一種黃酮苷,具有廣泛的藥理作用,臨床上主要用于心腦血管疾病的治療;秋水仙堿(colchicine)是從百合科植物秋水仙(Colchicumautumnale L.)的球莖中分離到的一種卓酚酮類生物堿,可用于治療痛風,對某些惡性腫瘤亦有較好的療效;喜樹堿(camptothecin,簡稱CPT)是從我國特有的天然植物喜樹(camptotheca acuminate)果實中分離出的一種具
[Abstract]:Trypsin is an animal-derived proteolytic enzyme with high specificity. A peptide bond that hydrolyzes only the carboxyl groups of L-arginine and L-lysine. Trypsin is a common activator of all trypsinogen (including its own precursor, trypsinogen). As an important digestive enzyme, trypsin is used in medicine. Food, industry and organic synthesis have been widely used and have been studied deeply. Nowadays, the focus is mainly on natural trypsin inhibitors. Alkaloids are a class of natural alkaloids containing nitrogen heterocycles, which are widely found in plants, most of which have important medical value. The interaction of three small alkaloid molecules with trypsin has been studied by means of the rapidly developed spectral analysis method in recent years. The three alkaloids selected in this paper are puerarin, colchicine and camptothecin. Puerarin is a flavonoid glycoside extracted from the roots of Pueraria lobata), which has a wide range of pharmacological effects, such as puerarin thomsoniiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiiii. Colchicine is an alkaloid isolated from the corm of Colchicum autumnale L., which can be used in the treatment of gout and has good curative effect on some malignant tumors. Camptothecin (CPT) was isolated from the fruit of camptotheca acuminate, a natural plant endemic to China.
