小鼠子宮內(nèi)膜中DBA、ECL和RCA-Ⅰ受體在胚泡植入過程中的表達(dá)狀況的實(shí)驗(yàn)研究

【摘要】:目的:胚胎著床是一個(gè)十分復(fù)雜的過程,是妊娠的關(guān)鍵所在,它涉及胚胎的發(fā)育分化、子宮內(nèi)膜的同步變化、胚泡對(duì)于子宮內(nèi)膜的黏附、侵入及子宮內(nèi)膜的包埋等環(huán)節(jié)。近年來(lái)的研究表明,胚泡著床過程不僅受到神經(jīng)內(nèi)分泌因素的調(diào)節(jié),而且還與胚泡和子宮內(nèi)膜產(chǎn)生的多種調(diào)節(jié)因子有關(guān),凝集素受體即是這些調(diào)節(jié)因子的一員。 凝集素受體是指分布于細(xì)胞表面的糖蛋白或糖脂。凝集素受體上具有能被凝集素特異識(shí)別的糖基。它們?cè)诩?xì)胞的生長(zhǎng)、分化、細(xì)胞之間的黏附及相互識(shí)別中起重要作用。為了探討子宮內(nèi)膜的凝集素受體與胚泡著床的關(guān)系,我們以早孕小鼠為研究對(duì)象,用親和細(xì)胞化學(xué)染色的方法檢測(cè)植入過程中子宮內(nèi)膜上皮相關(guān)凝集素受體分布狀況和變化規(guī)律,以明確與胚泡著床相關(guān)的凝集素受體并闡明它們?cè)谥仓械纳飳W(xué)效應(yīng),從而豐富生殖醫(yī)學(xué)中有關(guān)著床的理論,并為新型抗著床避孕方法的研究提供重要的實(shí)驗(yàn)依據(jù)。 研究方法:將健康育齡期雌性昆明小鼠行單側(cè)輸卵管結(jié)扎,2周后與正常有生育能力的雄鼠按2:1合籠交配,次日晨查見陰栓者定為孕第一天。分別于孕第4、6、7、8、9天頸椎脫臼處死孕鼠并立即剖腹取子宮(未孕側(cè)作為對(duì)照側(cè))。所得標(biāo)本用4%多聚甲醛固定,常規(guī)石蠟包埋,做厚度為6um的連續(xù)切片。采用SABC親和細(xì)胞化學(xué)法檢測(cè)DBA、ECL、RCA-Ⅰ三種凝集素的受體在著床期間的子宮內(nèi)膜的分布情況。 結(jié)果: 1 DBA染色結(jié)果:在懷孕一側(cè),DBA受體主要表達(dá)于胚胎滋養(yǎng)層細(xì)胞,子宮內(nèi)
[Abstract]:Objective: embryo implantation is a very complex process, which is the key of pregnancy. It involves the development and differentiation of embryo, the synchronous change of endometrium, the adhesion of embryo bubble to endometrium. Recent studies have shown that the implantation process of blastocyst is not only regulated by neuroendocrine factors, but also related to many regulatory factors produced by blastocyst and endometrium. Lectin receptors are one of these regulatory factors. Lectin receptors are glycoproteins or lipids distributed on the surface of cells. Lectin receptors have glycosyl groups that can be specifically recognized by lectins. They grow and differentiate in cells. In order to study the relationship between endometrial lectin receptor and blastocyst implantation, we studied early pregnant mice. The distribution and changes of endometrial epithelium-associated lectin receptors during implantation were detected by affinity cytochemical staining in order to identify the lectin receptors associated with blastocyst implantation and to elucidate their biological effects in implantation. It can enrich the theory of implantation in reproductive medicine and provide an important experimental basis for the study of new anti-implantation contraceptive methods. Methods: female Kunming mice of healthy reproductive age were treated with unilateral tubal ligation for 2 weeks to mate with normal fertile male mice in 2: 1 cage. The first day of pregnancy was found in the next morning. The pregnant rats were killed by cervical dislocations on the 4th day of pregnancy, and the uterus was immediately removed by laparotomy. The specimens were fixed with 4% polyformaldehyde and were routinely embedded in paraffin wax. SABC affinity cytochemistry was used to detect the distribution of three lectin receptors in the endometrium during implantation. Results:. 1 DBA staining: the DBA receptor was mainly expressed in trophoblast cells and uterus on the pregnant side.
