脈沖射頻對(duì)坐骨神經(jīng)慢性壓迫模型大鼠的痛覺過敏及脊髓背角NR2B亞基表達(dá)的作用

【摘要】:目的觀察脈沖射頻(PRF)對(duì)坐骨神經(jīng)慢性壓迫(CCI)后神經(jīng)病理性疼痛模型大鼠疼痛行為學(xué)、損傷坐骨神經(jīng)結(jié)組織病理學(xué)的影響,檢測(cè)脊髓背角NR2B亞基的表達(dá)。方法 48只Sprague-Dawley大鼠隨機(jī)等分為Sham-Sham(SS)組、Sham-PRF(SP)組、CCI-Sham(CS)組和CCI-PRF(CP)組。CS、CP組大鼠結(jié)扎右側(cè)坐骨神經(jīng)干,制作CCI模型;SS、SP組只分離坐骨神經(jīng)而不結(jié)扎。術(shù)后15 d,CP、SP組于坐骨神經(jīng)干結(jié)扎處行PRF,SS、CS組僅放置電極針不通電。造模前,造模后3 d、7 d、11 d、15 d,治療后1 d、3 d、7 d、11 d、15 d測(cè)量大鼠右后足機(jī)械縮足閾值(HWT);治療后15 d光鏡下行右側(cè)坐骨神經(jīng)干評(píng)分;Western blotting測(cè)定L4-6脊髓背角中NR2B亞基蛋白表達(dá)。結(jié)果造模后CS、CP組HWT較SS、SP組降低;治療后,CP組HWT高于CS組。光鏡下CP組較CS組神經(jīng)軸索直徑、髓鞘厚度明顯增加(P0.01)。CP組大鼠脊髓背角NR2B表達(dá)低于CS組(F=10.769,P0.05)。結(jié)論P(yáng)RF能降低CCI模型脊髓背角NR2B亞基表達(dá),修復(fù)受損神經(jīng),降低機(jī)械痛敏。
[Abstract]:Objective to observe the effect of pulse radiofrequency (PRF) on pain behavior and histology of injured sciatic nerve node in rats with neuropathic pain after chronic compression of (CCI), and to detect the expression of NR2B subunit in spinal dorsal horn. Methods 48 Sprague-Dawley rats were randomly divided into Sham-Sham (SS) group, Sham-PRF (SP) group, CCI-Sham (CS) group and CCI-PRF (CP) group. CS, CP group rats ligated the right sciatica trunk to make CCI model, SS,SP group only isolated the sciatica without ligating. On the 15th day after operation, PRF,SS,CS group only placed electrode needle without electricity in SP group. Before modeling, 3 days, 7 days, 15 days, 1 day, 3 days, 7 days and 11 days after treatment, the expression of NR2B subunit protein in the dorsal horn of spinal cord was measured by light microscope 15 days after (HWT); treatment.
