來(lái)源于肝纖維化環(huán)境內(nèi)皮祖細(xì)胞移植治療肝纖維化模型大鼠的逆轉(zhuǎn)作用

【摘要】:背景:理論上,從肝纖維化大鼠骨髓中提取經(jīng)過(guò)體內(nèi)病理環(huán)境"篩選"作用后的內(nèi)皮祖細(xì)胞,應(yīng)更能適應(yīng)肝纖維化微環(huán)境,移植入纖維化肝臟后應(yīng)更傾向于分化為成熟內(nèi)皮細(xì)胞,參與肝內(nèi)血管重建。目的:探討來(lái)源于大鼠肝纖維化體內(nèi)環(huán)境的骨髓源性內(nèi)皮祖細(xì)胞移植對(duì)大鼠肝纖維化的治療作用。方法:將28只Wistar大鼠隨機(jī)分為3組:正常組(n=8)皮下注射橄欖油,2次/周;模型組(n=10)皮下注射四氯化碳3 m L/kg,2次/周,首劑加倍,分別于第2,3,5周末尾靜脈輸注生理鹽水;實(shí)驗(yàn)組(n=10)皮下注射四氯化碳3 m L/kg,2次/周,首劑加倍,分別于第2,3,5周末尾靜脈輸注肝纖維化大鼠骨髓源性內(nèi)皮祖細(xì)胞懸液。注射6周末,進(jìn)行肝功能、凝血功能、肝組織相關(guān)蛋白及肝組織病理檢查。結(jié)果與結(jié)論:(1)肝組織病理:模型組肝組織呈廣泛而嚴(yán)重的肝細(xì)胞脂肪變性和壞死,門靜脈和中央靜脈周圍炎癥細(xì)胞浸潤(rùn),匯管區(qū)擴(kuò)大及大量纖維組織增生;與模型組相比,實(shí)驗(yàn)組肝細(xì)胞變性壞死、炎癥細(xì)胞浸潤(rùn)及纖維組織增生等均有所減輕;(2)肝組織相關(guān)蛋白:與正常組相比,模型組透明質(zhì)酸、層粘連蛋白和Ⅲ型前膠原、血管內(nèi)皮生長(zhǎng)因子、表皮生長(zhǎng)因子水平升高(P0.05);與模型組相比,實(shí)驗(yàn)組透明質(zhì)酸、層粘連蛋白和Ⅲ型前膠原水平明顯下降(P0.05),血管內(nèi)皮生長(zhǎng)因子、表皮生長(zhǎng)因子水平升高(P0.05);(3)肝功能、凝血功能:與正常組相比,模型組肝功能、凝血功能嚴(yán)重受損(P0.05);與模型組相比,實(shí)驗(yàn)組肝功能明顯改善(P0.05);(4)結(jié)果表明:來(lái)源于肝纖維化體內(nèi)環(huán)境的骨髓源性內(nèi)皮祖細(xì)胞移植能有效治療大鼠肝纖維化,其作用機(jī)制可能是通過(guò)促進(jìn)肝臟血管新生來(lái)實(shí)現(xiàn)的。
