端粒酶基因敲除小鼠行為學(xué)特征的增齡性變化研究

【摘要】:目的通過對端粒酶基因敲除小鼠衰老特點(diǎn)的初步認(rèn)識,為衰老領(lǐng)域的相關(guān)研究提供實(shí)驗(yàn)依據(jù)。方法分別選用2、3、5、7和9月齡的純合子與同月齡的野生型端粒酶基因敲除小鼠相對照,從老化度評分、Morris水迷宮實(shí)驗(yàn)和自主活動實(shí)驗(yàn)等方面觀察純合子小鼠行為活動的增齡性變化。結(jié)果與野生型小鼠比較,純合子小鼠隨月齡增加其老化度評分呈升高趨勢,且5、7和9月齡的老化度評分高于同齡野生型小鼠(P0.05);Morris水迷宮實(shí)驗(yàn)中,相比較野生型小鼠,純合子小鼠定位航行試驗(yàn)的逃避潛伏期隨月齡增加而有增長趨勢,且7和9月齡的逃避潛伏期長于同齡野生型小鼠(P0.05),而在空間探索試驗(yàn)的逃避潛伏期目標(biāo)象限時(shí)間隨月齡增加而呈下降趨勢,與野生型小鼠比較差異有統(tǒng)計(jì)學(xué)意義(P0.05);自主活動實(shí)驗(yàn)結(jié)果顯示,2、3和5月齡純合子小鼠固定時(shí)間內(nèi)自主活動次數(shù)與同齡野生型小鼠比較無顯著變化,而7和9月齡純合子小鼠與同齡野生型小鼠比較固定時(shí)間內(nèi)自主活動次數(shù)減少(P0.05)。結(jié)論與同齡野生型端粒酶基因敲除小鼠比較,純合子端粒酶基因敲除小鼠隨月齡增大,其行為活動出現(xiàn)明顯增齡性變化,表現(xiàn)出系列衰老特征,尤其7和9月齡衰老表現(xiàn)更為明顯。
[Abstract]:Objective to study the aging characteristics of telomerase knockout mice and to provide experimental basis for the related research in the field of aging. Methods the homozygotes from 2,3,5,7 and 9 months of age were compared with the wild type telomerase knockout mice of the same age, and the scores of aging degree were compared with those of the wild type telomerase knockout mice of the same age. Morris water maze test and autonomous activity test were used to observe the age-related changes of behavioral activity in homozygous mice. Results compared with wild-type mice, the aging degree scores of homozygote mice were higher than those of wild-type mice at 5, 7 and 9 months old (P0.05), and the aging degree score of homozygote mice was higher than that of wild-type mice at 5, 7 and 9 months old (P0.05). In the Morris water maze test, compared with wild mice, the escape latency of homozygote mice increased with the increase of month age, and the escape latency of 7 and 9 months old mice was longer than that of the same age wild type mice (P0.05), and the escape latency of homozygote mice was higher than that of wild type mice of the same age (P0.05). Compared with wild type mice, the escape latency target quadrant time in space exploration test decreased with the increase of age (P0.05). The results of autonomous activity test showed that there was no significant difference in the number of autonomous activities between the homozygote mice and the wild type mice of the same age in the fixed time of 2, 3 and 5 months old mice. However, the number of autonomous activities in homozygous mice at 7 and 9 months of age was lower than that in wild-type mice of the same age (P0.05). Conclusion compared with the wild type telomerase knockout mice of the same age, homozygote telomerase gene knockout mice have obvious age-related changes in their behavior with increasing age, showing a series of senescence characteristics, and a series of aging characteristics were observed in the homozygote telomerase gene knockout mice. Especially in July and September, the aging performance was more obvious.
