Chemerin與炎癥、動脈粥樣硬化的相關(guān)性研究

【摘要】:目的:1、比較三組Wistar大鼠血漿Chemerin、HS-CRP、IL-6、TNF-p水平變化;觀察三組Wistar大鼠主動脈Chemerin基因和Chemerin R基因表達情況。2、比較正常對照組和冠心病組之間血漿Chemerin水平,分析血漿Chemerin水平與臨床一般、生化資料的相關(guān)性。方法:1、將健康雄性Wistar大鼠82只,分為對照組(D)、辛伐他汀治療組(X)及高脂組(G)。喂養(yǎng)14周處死,行心臟采血、主動脈弓HE染色及分離胸主動脈和腹主動脈。2、ELISA法檢測三組大鼠血漿Chemerin、HS-CRP、IL-6、TNF-β水平變化,采用半定量RT-PCR技術(shù)檢測三組大鼠主動脈Chemerin-Chemerin R mRNA水平。3、用雙抗體夾心法檢測36例正常人和75例冠心病患者血漿Chemerin水平,同時測定受試者的體重指數(shù)(BMI)、腰臀比(WHR)、血壓,檢測血糖、血脂、超敏C反應(yīng)蛋白(HS-CRP)等指標。結(jié)果:1、高脂組大鼠動脈粥樣硬化(AS)形成良好。2、三組大鼠間血漿Chemerin、HS-CRP、IL-6、TNF-β水平差異具有統(tǒng)計學(xué)意義,均呈高脂組辛伐他汀治療組對照組,P均0.01;高脂組大鼠血漿Chemerin與HS-CRP、IL-6、TNF-β呈正相關(guān)(r=0.804、0.614、0.821,P0.01),辛伐他汀治療組與高脂組相似。3、三組大鼠間主動脈Chemerin-Chemerin R mRNA表達水平差異具有統(tǒng)計學(xué)意義,呈高脂組辛伐他汀治療組對照組,P均0.01;高脂組、辛伐他汀治療組Chemerin-Chemerin R mRNA表達水平與HS-CRP、IL-6、TNF-β相關(guān)性和相應(yīng)組血漿Chemerin與HS-CRP、IL-6、TNF-β水平相關(guān)性相似。4、冠心病組與正常對照組血漿Chemerin水平差異具有統(tǒng)計學(xué)意義(P0.01),且與冠脈病變支數(shù)呈正相關(guān)(r=0.423,P0.05);在正常對照組,女性較男性有較高血漿Chemerin水平(P0.05);冠心病患者血漿Chemerin水平與HS-CRP、LDL呈正相關(guān),r=0.512、0.246,P0.05;在校正年齡、性別、BMI、WHR、 BP后,血漿Chemerin與HS-CRP仍呈正相關(guān)(r=0.264,P0.05)。以Chernerin為因變量,作多元Logistic回歸分析,有SBP、WHR、HS-CRP進入方程(P0.05)。結(jié)論:1、大鼠血漿Chemerin水平、主動脈Chemerin-Chemerin R基因mRNA表達水平均與炎癥有明顯相關(guān)性,而AS是一種慢性炎癥過程,Chemerin與AS的形成可能具有某種內(nèi)在聯(lián)系。2、冠心病組Chemerin明顯高于正常對照組,且與冠狀動脈病變指數(shù)呈正相關(guān),冠心病主要的病理是AS,提示這種新的脂肪因子可能在冠心病及其相關(guān)心血管疾病的病理生理機制中發(fā)揮作用。
[Abstract]:Objective: 1. To compare the changes of plasma Chemerin,HS-CRP,IL-6,TNF-p levels in three groups of Wistar rats. The expression of Chemerin gene and Chemerin R gene in aorta of three groups of Wistar rats were observed. 2. The plasma Chemerin level was compared between normal control group and coronary heart disease group, and the correlation between plasma Chemerin level and clinical and biochemical data was analyzed. Methods: 1. 82 healthy male Wistar rats were divided into control group, (D), simvastatin treatment group, (X) group and hyperlipidemic group (G). After 14 weeks of feeding, the rats were sacrificed, the blood was collected from the heart, the aortic arch was stained with HE, the thoracic aorta and abdominal aorta were separated, and the plasma Chemerin,HS-CRP,IL-6,TNF- 尾 levels in the three groups were detected by Elisa. The levels of Chemerin-Chemerin R mRNA in aorta of three groups of rats were measured by semi-quantitative RT-PCR technique. The plasma Chemerin levels of 36 normal subjects and 75 patients with coronary heart disease were detected by double antibody sandwich method. The body mass index (BMI),) was also measured. Blood pressure, blood glucose, blood lipid and high sensitive C reactive protein (HS-CRP) were measured in waist hip ratio (WHR),). Results: 1. The formation of atherosclerosis (AS) was good in the high fat group. 2There was significant difference in plasma Chemerin,HS-CRP,IL-6,TNF- 尾 level among the three groups. P both 0.01; There was a positive correlation between plasma Chemerin and HS-CRP,IL-6,TNF- 尾 in hyperlipidemia group (r = 0.804 / 0. 6140. 821 / P0.01), while simvastatin treatment was similar to that in hyperlipidemia group. The expression of Chemerin-Chemerin R mRNA in aorta of the three groups was significantly different (P < 0.01). The expression of Chemerin-Chemerin R mRNA in hyperlipidemia group and simvastatin group was similar to that of HS-CRP,IL-6,TNF- 尾 and plasma Chemerin and HS-CRP,IL-6,TNF- 尾 levels. There was significant difference in plasma Chemerin level between coronary heart disease group and normal control group (P0.01), and there was a positive correlation with the number of coronary artery disease branches (r = 0.423 P 0.05). In the normal control group, the plasma Chemerin level of female was higher than that of male (P0.05), the plasma Chemerin level of coronary heart disease patients was positively correlated with HS-CRP,LDL (r = 0.512, 0.246, P 0.05). After adjusting for age, sex and BMI,WHR, BP, plasma Chemerin was still positively correlated with HS-CRP (r = 0.264, P 0.05). With Chernerin as dependent variable, multiple Logistic regression analysis was made. SBP,WHR,HS-CRP entered the equation (P0.05). Conclusion: 1. The levels of Chemerin in plasma and mRNA expression of Chemerin-Chemerin R gene in aorta are significantly correlated with inflammation, while AS is a chronic inflammatory process, and the formation of Chemerin and AS may have some internal relationship. 2. The Chemerin of coronary heart disease group was significantly higher than that of normal control group, and had positive correlation with coronary artery disease index. The main pathology of coronary heart disease was AS,. These results suggest that this new fat factor may play a role in the pathophysiological mechanism of coronary heart disease and its related cardiovascular diseases.
