氫醌對骨髓間充質(zhì)干細(xì)胞PARP-1表達(dá)的影響

【摘要】:目的探討氫醌(HQ)對骨髓間充質(zhì)干細(xì)胞(BMSCs)PARP-1基因表達(dá)的影響。方法 2015年1月—2015年3月在東莞市環(huán)境醫(yī)學(xué)重點(diǎn)實(shí)驗(yàn)室以BMSCs為對照組,以PARP-1沉默BMSCs為處理組,磷酸鹽緩沖液(PBS)溶解HQ,分別以0、2.5、5.0、10.0、20.0和40.0μmol/L HQ染毒兩組細(xì)胞,應(yīng)用實(shí)時(shí)熒光定量-聚合酶鏈反應(yīng)檢測兩組細(xì)胞PARP-1基因表達(dá)的改變。結(jié)果與PBS組對比,2.5、5.0、10.0、20.0、40.0μmol/L HQ染毒24 h后,BMSCs細(xì)胞PARP-1表達(dá)量分別為1.39倍、1.65倍(P0.05)、1.69倍(P0.05)、1.76倍(P0.05)和1.5倍(P0.05);沉默細(xì)胞PARP-1表達(dá)量分別為1.37倍(P0.05)、1.58倍(P0.05)、1.84倍(P0.05)、2.15倍(P0.05)和1.93倍(P0.05);與BMSCs細(xì)胞相比,沉默細(xì)胞中PARP-1表達(dá)水平下降了78%(P0.05)差異均無統(tǒng)計(jì)學(xué)意義。結(jié)論兩組細(xì)胞PARP-1表達(dá)水平隨HQ劑量增加先升后降,PARP-1沉默能增加BMSCs對HQ的敏感性。
[Abstract]:Objective to investigate the effect of hydroquinone (HQ) on (BMSCs) PARP-1 gene expression in bone marrow mesenchymal stem cells. Methods from January 2015 to March 2015, BMSCs was used as control group and PARP-1 silencing BMSCs as treatment group. Phosphate buffer (PBS) dissolved HQ, in Dongguan Environmental Medicine key Laboratory. The changes of PARP-1 gene expression in the two groups were detected by real-time fluorescence quantification-polymerase chain reaction (FQ-PCR). Results compared with PBS group, the expression of PARP-1 in BMSCs cells was 1.39 times, 1.65 times (P0.05), 1.69 times (P0.05), respectively. 1.76 times (P0.05) and 1.5 times (P0.05); The expression of PARP-1 in silencing cells was 1.37 times (P0.05), 1.58 times (P0.05), 1.84 times (P0.05), 2.15 times (P0.05) and 1.93 times (P0.05) respectively. Compared with BMSCs cells, the level of PARP-1 expression in silencing cells decreased by 78% (P0.05). Conclusion the expression of PARP-1 in the two groups increased first with the increase of HQ dose and then decreased. PARP-1 silencing could increase the sensitivity of BMSCs to HQ.
