弗氏枸櫞酸桿菌CF74的T6SS影響FliC的表達(dá)和分泌

【摘要】:目的研究弗氏枸櫞酸桿菌CF74的T6SS基因組島功能。方法構(gòu)建弗氏枸櫞酸桿菌CF74的T6SS基因組島的精確缺失突變株(CF74ΔT6SS)。對(duì)弗氏枸櫞酸桿菌CF74和其突變株CF74ΔT6SS的全菌蛋白和上清蛋白進(jìn)行提取,并進(jìn)行2-DE分析。結(jié)果在CF74ΔT6SS全菌蛋白中,有4個(gè)蛋白表達(dá)上調(diào),42個(gè)蛋白表達(dá)下調(diào),并且Fli C的表達(dá)下調(diào)十分明顯;在CF74ΔT6SS的培養(yǎng)上清中,有27個(gè)蛋白表達(dá)上調(diào),36個(gè)蛋白表達(dá)下調(diào),并且Fli C、Flg K和Fli D的表達(dá)下調(diào)十分明顯。結(jié)論弗氏枸櫞酸桿菌CF74的T6SS可以影響Fli C蛋白的表達(dá)和分泌。
[Abstract]:Objective to study the T6SS genomic island function of Citrobacter flexneri CF74. Methods the CF74 螖 T6SS of the T6SS genomic island of Citrobacter flexneri CF74 was constructed. The whole and supernatant proteins of Citrobacter flexneri CF74 and its mutant CF74 螖 T6SS were extracted and analyzed by 2-DE. Results in the whole bacterial protein of CF74 螖 T6SS, 4 proteins were up-regulated, 42 proteins were down-regulated, and the expression of Fli C was down-regulated. In the culture supernatant of CF74 螖 T6SS, 27 proteins were up-regulated, 36 proteins were down-regulated, and the expression of Fli Cnflg K and Fli D were down-regulated. Conclusion T6SS of Citrobacter flexneri CF74 can affect the expression and secretion of Fli C protein.
