溶血磷脂酸對(duì)缺氧培養(yǎng)大鼠乳鼠心室肌細(xì)胞動(dòng)作電位及L-型鈣電流的影響

【摘要】:目的: 本實(shí)驗(yàn)通過觀察外源性溶血磷脂酸(LPA)對(duì)缺氧條件下原代培養(yǎng)大鼠乳鼠心室肌細(xì)胞動(dòng)作電位及L-型鈣電流的影響,探討LPA對(duì)缺氧培養(yǎng)大鼠乳鼠心室肌細(xì)胞的作用。 方法: 應(yīng)用全細(xì)胞膜片鉗技術(shù),觀察缺氧培養(yǎng)對(duì)大鼠乳鼠心室肌細(xì)胞動(dòng)作電位的影響;觀察不同劑量LPA對(duì)缺氧培養(yǎng)大鼠乳鼠心室肌細(xì)胞動(dòng)作電位及L-型鈣電流的影響。 結(jié)果: 缺氧培養(yǎng)后的心室肌細(xì)胞與正常細(xì)胞相比搏動(dòng)頻率減慢、搏動(dòng)幅度減弱伴有節(jié)律不齊和部分停搏,并隨培養(yǎng)時(shí)間延長(zhǎng)程度加重。觀察到缺氧培養(yǎng)不同時(shí)間后,動(dòng)作電位的時(shí)程隨缺氧時(shí)間延長(zhǎng)而縮短,給予外源性LPA可延長(zhǎng)動(dòng)作電位時(shí)程;缺氧培養(yǎng)可使大鼠乳鼠心室肌細(xì)胞L-型鈣電流的電流幅度明顯減小給予外源性LPA使其幅值升高。 結(jié)論: 1.缺氧培養(yǎng)大鼠乳鼠心室肌細(xì)胞與正常心室肌細(xì)胞相比,搏動(dòng)頻率減慢、搏動(dòng)幅度減弱并伴有節(jié)律不齊和部分停搏; 2.缺氧培養(yǎng)條件下大鼠乳鼠心室肌細(xì)胞動(dòng)作電位各參數(shù)減小,且具有時(shí)間依從性;給予外源性LPA可延長(zhǎng)動(dòng)作電位時(shí)程; 3.缺氧培養(yǎng)可使大鼠乳鼠心室肌細(xì)胞L-型鈣電流減小,且具有時(shí)間依從性; 4.LPA能夠增強(qiáng)缺氧培養(yǎng)條件下大鼠乳鼠心室肌細(xì)胞L-型鈣電流的幅值,且具有劑量依賴性。
[Abstract]:Objective: to observe the effects of exogenous lysophosphatidic acid (LPA) on action potential and L-type calcium current in cultured rat ventricular myocytes under hypoxia. To investigate the effect of LPA on ventricular myocytes cultured in neonatal rats. Methods: the effects of hypoxia culture on action potential of ventricular myocytes in neonatal rats and the effects of different doses of LPA on action potential and L-type calcium current of ventricular myocytes in neonatal rats were observed by whole-cell patch clamp technique. Results: compared with the normal cells, the ventricular myocytes cultured under hypoxia had slower pulsatile frequency, lower pulsation amplitude, irregular rhythm and partial arrest, and the degree of pulsation increased with the prolongation of culture time. It was observed that the duration of action potential was shortened with the prolongation of anoxic time after hypoxia culture. Exogenous LPA could prolong the duration of action potential. Hypoxia culture significantly reduced the amplitude of L-type calcium current in neonatal rat ventricular myocytes and increased the amplitude of L- type calcium current by exogenous LPA. Conclusion: 1. Compared with normal ventricular myocytes, hypoxic cultured rat ventricular myocytes had slower pulsatile frequency, lower pulsatile amplitude, irregular rhythm and partial arrest; 2. The parameters of action potential of ventricular myocytes in neonatal rats were decreased in hypoxic culture and had time compliance; exogenous LPA prolonged the duration of action potential; 3. Hypoxia culture could decrease the L-type calcium current in neonatal rat ventricular myocytes, and 4.LPA could enhance the amplitude of L-type calcium current in neonatal rat ventricular myocytes in a dose-dependent manner.
