續(xù)骨丹外敷療法對大鼠脛骨缺損模型血清BMP-2的影響

【摘要】:為觀察續(xù)骨丹外敷療法對大鼠脛骨缺損模型血清骨形態(tài)發(fā)生蛋白-2(BMP-2)表達的影響。給70只SD大鼠右側脛骨制備1個4 mm缺損,用0.8 mm克氏針內固定。造模1個月后符合標準的大鼠隨機分為實驗組(藥物治療組)、對照組a(脈沖磁場治療組)、對照組b(面粉干擾組)和空白組。在干預前、2周、4周后對大鼠右脛骨行X線檢查,然后處死動物,收獲骨樣本,做蘇木素-伊紅(HE)染色和血清BMP-2檢測。干預前,實驗組、對照組a、對照組b和空白組,4組大鼠血清BMP-2的含量沒有明顯差異;干預至第2周時,4組大鼠血清BMP-2的含量與干預前比較均下降,但組間差異均無統(tǒng)計學意義(p0.05);干預4周后,實驗組大鼠血清BMP-2的含量高于對照組a、b和空白組,差異均有統(tǒng)計學意義(p0.05),對照組a中BMP-2的含量高于對照組b和空白組,差異均有統(tǒng)計學意義(p0.05),對照組b與空白組比較,差異無統(tǒng)計學意義(p0.05)。4組大鼠血清BMP-2的含量在干預前、2周和4周后的變化趨勢是:實驗組和對照組a先降后升,對照組b和空白組一直下降。續(xù)骨丹外敷療法和脈沖電磁場療法在實驗干預至第4周時均能上調骨缺損大鼠BMP-2的表達,有利于骨缺損的愈合,但續(xù)骨丹外敷療法對BMP-2的調節(jié)作用上明顯優(yōu)于脈沖電磁場。
[Abstract]:To observe the effect of external application of Xuguodan on the expression of serum bone morphogenetic protein-2 (BMP-2) in rat tibial defect model. A 4 mm defect was prepared from the right tibia of 70 SD rats and fixed with 0.8 mm Kirschner needle. One month later, the rats who met the standard were randomly divided into three groups: experimental group (drug treatment group), control group a (pulse magnetic field treatment group), control group b (flour interference group) and blank group. The right tibia of rats was examined by X-ray before, 2 weeks and 4 weeks after intervention, and then the animals were killed, bone samples were harvested, and hematoxylin eosin (HE) staining and serum BMP-2 detection were performed. Before intervention, there was no significant difference in serum BMP-2 content among experimental group, control group b and blank group. At the second week of intervention, the levels of serum BMP-2 in the four groups decreased compared with those before the intervention, but there was no significant difference between the four groups (p0.05). After 4 weeks of intervention, the level of serum BMP-2 in experimental group was higher than that in control group and blank group, the difference was statistically significant (p0.05). The content of BMP-2 in control group a was higher than that in control group and blank group. The difference was statistically significant (p0.05), but there was no significant difference between control group b and blank group (p0.05). The content of serum BMP-2 in the 4 groups was before intervention, and no significant difference was found between the control group and the control group (p0.05). After 2 and 4 weeks, the change trend was: a of the experimental group and the control group decreased first and then increased, and that of the control group b and the blank group decreased all the time. The expression of BMP-2 in rats with bone defect could be upregulated by external application of Xuguodan and pulsed electromagnetic field therapy at the 4th week, which was beneficial to the healing of bone defect. But the effect of external application of Xiegudan on the regulation of BMP-2 was obviously superior to that of pulsed electromagnetic field.
