人類(lèi)臍帶血MSC和骨髓MSC分泌細(xì)胞因子的對(duì)比研究

【摘要】:目的應(yīng)用人類(lèi)臍帶血和健康人骨髓這兩種材料,分別培養(yǎng)出間充質(zhì)干細(xì)胞,然后比較兩種細(xì)胞分泌支持造血調(diào)控細(xì)胞因子的差異,為體外高效造血干細(xì)胞擴(kuò)增體系的確立提供參考,明確臍帶血來(lái)源的間充質(zhì)干細(xì)胞的支持造血的能力。 方法用ELISA方法,測(cè)定兩種細(xì)胞分泌的干細(xì)胞生長(zhǎng)因子(Stem cell factor , SCF),血小板生成素(Thrombopoietin , TPO)和白細(xì)胞介素6(Interleukin-6,IL-6)的濃度,并且進(jìn)行對(duì)比。 結(jié)果在體外培養(yǎng)過(guò)程中,臍帶血及骨髓中的間充質(zhì)干細(xì)胞均可表達(dá)干細(xì)胞生長(zhǎng)因子(Stem cell factor , SCF),血小板生成素(Thrombopoietin , TPO)和白細(xì)胞介素6(Interleukin-6,IL-6)。但是相同生長(zhǎng)條件下,臍帶血及骨髓中的間充質(zhì)干細(xì)胞分泌的因子濃度不同。人臍血基質(zhì)細(xì)胞分泌SCF的量(575.7±40.2pg/ml)高于骨髓基質(zhì)細(xì)胞(482.2±17.7pg/ml),而人臍血基質(zhì)細(xì)胞分泌IL-6 (469.7±27.2pg/ml)的量低于同期培養(yǎng)的骨髓基質(zhì)細(xì)胞(935.19±35.4pg/ml),人臍血基質(zhì)細(xì)胞TPO分泌的量(195.5±7.3pg/ml)與骨髓基質(zhì)細(xì)胞(172.2±9.3pg/ml)相近。
[Abstract]:Objective to culture mesenchymal stem cells (MSCs) from human umbilical cord blood (UCB) and healthy human bone marrow (BMSCs), and compare the difference of cytokines secreted by two kinds of cells in supporting hematopoietic regulation. To provide a reference for the establishment of an in vitro high efficient hematopoietic stem cell amplification system and to clarify the ability of umbilical cord blood derived mesenchymal stem cells to support hematopoiesis. Methods ELISA assay was used to detect the concentrations of (Stem cell factor, SCF), thrombopoietin (Thrombopoietin, TPO) and interleukin 6 (Interleukin-6,IL-6) secreted by two kinds of stem cell growth factor (Stem cell factor, SCF), and the results were compared. Results in vitro, mesenchymal stem cells in cord blood and bone marrow could express stem cell growth factor (Stem cell factor, SCF), thrombopoietin (Thrombopoietin, TPO) and interleukin 6 (Interleukin-6,IL-6). However, under the same growth conditions, the concentration of factors secreted by mesenchymal stem cells in cord blood and bone marrow varied. The amount of SCF secreted by human umbilical cord blood stromal cells (575.7 鹵40.2pg/ml) was higher than that of bone marrow stromal cells (482.2 鹵17.7pg/ml). However, the amount of IL-6 secreted by human umbilical cord blood stromal cells (469.7 鹵27.2pg/ml) was lower than that of bone marrow stromal cells cultured at the same time (935.19 鹵35.4pg/ml). The amount of TPO secreted by human umbilical cord blood stromal cells (195.5 鹵7.3pg/ml) was similar to that of bone marrow stromal cells (172.2 鹵9.3pg/ml).
