人類臍帶血MSC和骨髓MSC分泌細胞因子的對比研究

【摘要】:目的應用人類臍帶血和健康人骨髓這兩種材料,分別培養(yǎng)出間充質干細胞,然后比較兩種細胞分泌支持造血調控細胞因子的差異,為體外高效造血干細胞擴增體系的確立提供參考,明確臍帶血來源的間充質干細胞的支持造血的能力。 方法用ELISA方法,測定兩種細胞分泌的干細胞生長因子(Stem cell factor , SCF),血小板生成素(Thrombopoietin , TPO)和白細胞介素6(Interleukin-6,IL-6)的濃度,并且進行對比。 結果在體外培養(yǎng)過程中,臍帶血及骨髓中的間充質干細胞均可表達干細胞生長因子(Stem cell factor , SCF),血小板生成素(Thrombopoietin , TPO)和白細胞介素6(Interleukin-6,IL-6)。但是相同生長條件下,臍帶血及骨髓中的間充質干細胞分泌的因子濃度不同。人臍血基質細胞分泌SCF的量(575.7±40.2pg/ml)高于骨髓基質細胞(482.2±17.7pg/ml),而人臍血基質細胞分泌IL-6 (469.7±27.2pg/ml)的量低于同期培養(yǎng)的骨髓基質細胞(935.19±35.4pg/ml),人臍血基質細胞TPO分泌的量(195.5±7.3pg/ml)與骨髓基質細胞(172.2±9.3pg/ml)相近。
[Abstract]:Objective to culture mesenchymal stem cells (MSCs) from human umbilical cord blood (UCB) and healthy human bone marrow (BMSCs), and compare the difference of cytokines secreted by two kinds of cells in supporting hematopoietic regulation. To provide a reference for the establishment of an in vitro high efficient hematopoietic stem cell amplification system and to clarify the ability of umbilical cord blood derived mesenchymal stem cells to support hematopoiesis. Methods ELISA assay was used to detect the concentrations of (Stem cell factor, SCF), thrombopoietin (Thrombopoietin, TPO) and interleukin 6 (Interleukin-6,IL-6) secreted by two kinds of stem cell growth factor (Stem cell factor, SCF), and the results were compared. Results in vitro, mesenchymal stem cells in cord blood and bone marrow could express stem cell growth factor (Stem cell factor, SCF), thrombopoietin (Thrombopoietin, TPO) and interleukin 6 (Interleukin-6,IL-6). However, under the same growth conditions, the concentration of factors secreted by mesenchymal stem cells in cord blood and bone marrow varied. The amount of SCF secreted by human umbilical cord blood stromal cells (575.7 鹵40.2pg/ml) was higher than that of bone marrow stromal cells (482.2 鹵17.7pg/ml). However, the amount of IL-6 secreted by human umbilical cord blood stromal cells (469.7 鹵27.2pg/ml) was lower than that of bone marrow stromal cells cultured at the same time (935.19 鹵35.4pg/ml). The amount of TPO secreted by human umbilical cord blood stromal cells (195.5 鹵7.3pg/ml) was similar to that of bone marrow stromal cells (172.2 鹵9.3pg/ml).
