手足綜合征大鼠模型血漿細(xì)胞因子的變化

【摘要】:目的建立大鼠手足綜合征模型,探討手足綜合征發(fā)病過程中相關(guān)炎性因子的水平及發(fā)病機(jī)制。方法運(yùn)用卡培他濱制作手足綜合征大鼠模型,分為給藥1周期的A組和給藥兩周期的B組(連續(xù)給藥7d,間隔3d,10d為一個(gè)周期),分別模擬不同給藥劑量下發(fā)生的手足綜合征,留取大鼠足跖部圖像資料及血漿標(biāo)本,檢測血漿中親膽堿能神經(jīng)元因子(CNTF)、血小板衍生的內(nèi)皮細(xì)胞生長因子(PD-ECGF)、白介素β(IL-β)、白介素10(IL-10)、中性粒細(xì)胞趨化因子(CINC-3)水平。結(jié)果與正常大鼠相比,1周期模型大鼠PD-ECGF,CINC-3,CNTF,IL-10,IL-β水平均升高(P0.01);而1周期造模未成功大鼠、模型大鼠停藥1周后各細(xì)胞因子水平與正常大鼠相比的差異無統(tǒng)計(jì)學(xué)意義(P0.05)。與正常大鼠相比,2周期模型大鼠及模型大鼠停藥1周后CINC-3,CNTF水平明顯升高(P0.01);而余各因子在各組間水平差異無統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論在短期給藥過程中,HFS的發(fā)生主要與廣泛發(fā)生的炎性損傷有關(guān),而在較長時(shí)間的給藥后,廣泛的炎性反應(yīng)趨于控制,而主要反應(yīng)為相關(guān)炎性細(xì)胞的趨化及神經(jīng)元的損傷。
[Abstract]:Objective to establish a rat model of hand and foot syndrome and to explore the level and pathogenesis of inflammatory factors in the pathogenesis of hand and foot syndrome. Methods the rat model of hand and foot syndrome was established by capecitabine. The rats were divided into two groups: group A (1 cycle) and group B (2 cycles) (7 days of continuous administration of capecitabine at intervals of 3 days and 10 days as a cycle). The paw and metatarsal images of rats and plasma samples were used to detect the endothelium-derived endothelial growth factor (PD-ECGF) derived from (CNTF), platelet-derived cholinergic neuron factor (CNTF),). Levels of interleukin 尾 (IL- 尾), interleukin 10 (IL-10) and neutrophil chemokine (CINC-3). Results compared with normal rats, the level of PD-ECGF,CINC-3,CNTF,IL-10,IL- 尾 increased in 1 cycle model rats (P0.01). However, the level of cytokines in the model rats was not significantly different from that in the normal rats after 1 week of withdrawal (P0.05). Compared with normal rats, the level of CINC-3,CNTF increased significantly in 2-cycle model rats and model rats after 1 week of withdrawal (P0.01), while the other factors were not significantly different among the groups (P0.05). Conclusion during the short-term administration of HFS, the occurrence of HFS is mainly related to the extensive inflammatory injury, and the extensive inflammatory response tends to be controlled after a long time of administration, and the main response is the chemotaxis of the related inflammatory cells and the injury of neurons.
