人胎盤間充質(zhì)樣干細(xì)胞的分離、培養(yǎng)及其生物學(xué)特性分析

【摘要】:從組織塊中分離純化成體干細(xì)胞具有一定的困難,其他細(xì)胞的混雜成為獲取高純度組織干細(xì)胞的一個技術(shù)瓶頸.以人體胎盤為研究對象,在分散組織細(xì)胞的基礎(chǔ)上,通過不同時間梯度貼壁方法分離純化了人胎盤間充質(zhì)樣干細(xì)胞,并與常規(guī)的直接貼壁方法作了比較,同時分析了時間梯度貼壁方法分選細(xì)胞的增殖能力、多分化潛能以及免疫原性.研究結(jié)果表明,時間梯度貼壁法獲得的細(xì)胞具有較強(qiáng)的增殖能力.直接貼壁法得到的細(xì)胞不具有分化潛能,而時間梯度貼壁法獲得的細(xì)胞具有向成骨細(xì)胞和成脂肪細(xì)胞方向分化的能力.通過對免疫原性基因檢測的分析,表明時間梯度貼壁法獲得的細(xì)胞表達(dá)HLA-Ⅰ,但不表達(dá)HLA-Ⅱ.
[Abstract]:It is difficult to isolate and purify adult stem cells from tissue blocks, and the mixing of other cells has become a technical bottleneck in obtaining high purity tissue stem cells. Human placenta mesenchymal stem cells were isolated and purified by different time gradient adherent methods on the basis of dispersed tissue cells. At the same time, the proliferative ability, multi-differentiation potential and immunogenicity of the cells were analyzed by time gradient adherent method. The results showed that the cells obtained by time gradient adhesion method had strong proliferative ability. The cells obtained by direct adherent method have no differentiation potential, but the cells obtained by time gradient adhesion method have the ability to differentiate into osteoblasts and adipoblasts.
