CIK對(duì)SGC-7901胃癌移植瘤小鼠模型體內(nèi)抗腫瘤作用的實(shí)驗(yàn)研究

【摘要】:目的探討細(xì)胞因子誘導(dǎo)殺傷細(xì)胞(CIK)對(duì)胃癌移植瘤小鼠模型的抗腫瘤作用并探討其作用機(jī)制。方法體內(nèi)實(shí)驗(yàn)選取60只C57BL/6小鼠分為模型對(duì)照組、5-Fu組、CIK大劑量、中劑量、小劑量組和正常對(duì)照組(n=10),分別治療周期為4周,比較各組胃癌小鼠治療期間移植瘤生長(zhǎng)情況,測(cè)定各組小鼠血清和腫瘤組織MMP-9、VEGF及bFGF水平。結(jié)果 1)CIK中劑量組小鼠瘤體積于治療后1周、2周、3周和4周時(shí)均較CIK小劑量組顯著減少(P0.05),CIK大劑量組小鼠瘤體積治療后1周、2周、3周和4周時(shí)均較CIK中劑量組顯著減少(P0.05);2)治療4周時(shí)CIK中劑量組小鼠血清MMP-9、VEGF和bFGF水平均較小劑量組小鼠顯著降低(P0.05),CIK大劑量組小鼠血清MMP-9、VEGF和bFGF水平均較中劑量組小鼠顯著降低(P0.05);3)治療4周時(shí)CIK中劑量組小鼠移植瘤組織MMP-9、VEGF和bFGF mRNA和蛋白水平均較小劑量組小鼠顯著降低(P0.05),CIK大劑量組小鼠移植瘤組織MMP-9、VEGF和bFGF mRNA和蛋白水平均較中劑量組小鼠顯著降低(P0.05)。結(jié)論細(xì)胞因子誘導(dǎo)殺傷細(xì)胞可顯著抑制SGC-7901胃癌小鼠模型瘤組織的生長(zhǎng),并且降低血液及癌組織中MMP-9、VEGF和bFGF的表達(dá)。
[Abstract]:Objective to investigate the antitumor effect of cytokine induced killer cell (CIK) on gastric cancer transplanted tumor mice and its mechanism. Methods Sixty C57BL/6 mice were divided into three groups: model control group, 5-Fu group, high dose, middle dose of CIK, small dose group and normal control group (n = 10). The treatment period was 4 weeks, respectively. The growth of transplanted tumor was compared during the treatment of gastric cancer mice in each group. The levels of MMP-9,VEGF and bFGF in serum and tumor tissue of each group were measured. Results 1) the tumor volume in CIK medium dose group was significantly lower than that in CIK small dose group at 1 week, 2 weeks, 3 weeks and 4 weeks after treatment (P0.05). The tumor volume in), CIK group was significantly lower than that in CIK medium dose group at 1 week, 2 weeks, 3 weeks and 4 weeks after treatment. The levels of serum MMP-9,VEGF and bFGF in the CIK medium dose group were significantly lower than those in the low dose group at 4 weeks after treatment (P0.05) (P0.05) the serum MMP-9,VEGF and bFGF levels in the high dose group were significantly lower than those in the middle dose group (P0.05); 3) at 4 weeks after treatment, the level of CIK in the medium dose group was significantly lower than that in the middle dose group (P0.05). The levels of MMP-9,VEGF and bFGF mRNA and protein in transplanted tumor tissue of mice in the group B were significantly lower than those in the group B (P0.05). The levels of MMP-9,VEGF and bFGF mRNA and protein in the transplanted tumor tissue of the high-dose), CIK group were significantly lower than those of the mice in the middle dose group (P0.05). Conclusion Killer cells induced by cytokines can significantly inhibit the growth of SGC-7901 gastric cancer mice model tumor tissues and decrease the expression of MMP-9,VEGF and bFGF in blood and cancer tissues.
