【摘要】：目的研究復(fù)方苦參注射液對(duì)乳腺癌模型大鼠的改善作用。方法按照隨機(jī)數(shù)字法選雄性SD大鼠15只為正常組,余采用腹腔注射甲基亞硝基脲(MNU)復(fù)制大鼠乳腺癌模型。按隨機(jī)數(shù)字表法將造模成功大鼠隨機(jī)分為5組:模型組,對(duì)照組,大中小3個(gè)劑量實(shí)驗(yàn)組,各15只。從造模成功第2天開始,實(shí)驗(yàn)組腹腔注射復(fù)方苦參注射液80,40,20 m L·kg~(-1),模型組灌胃三苯氧胺混懸液40 m L·kg~(-1),對(duì)照組和正常組大鼠灌胃生理鹽水4 m L·kg~(-1),每組均1次/天。干預(yù)9周后,用酶聯(lián)免疫吸附法測(cè)定大鼠血清中的雌二醇(E2)與存活素含量、乳腺組織內(nèi)的細(xì)胞增殖抗原標(biāo)記物(Ki67)、抑癌基因p53(P53)及組織和血清的程序性細(xì)胞死亡因子4(PDCD4)水平。結(jié)果正常組、對(duì)照組、小中大3個(gè)劑量實(shí)驗(yàn)組和模型組的E2含量分別為(36.92±1.28),(70.09±1.91),(64.40±2.77),(40.49±2.17),(40.49±2.39),(49.56±1.27)ng·L~(-1);這6組的Ki67分別為(15.72±0.76),(70.25±1.59),(65.41±1.33),(21.03±1.50),(21.30±1.10),(40.91±1.85)ng·L~(-1);這6組的P53分別為(10.63±0.89),(50.41±1.73),(45.04±2.09),(11.83±1.29),(12.18±1.48),(28.37±1.66)ng·L~(-1);這6組的存活素分別為(5.95±0.46),(63.91±1.96),(30.85±1.63),(16.96±0.53),(16.58±0.95),(25.36±1.51)ng·L~(-1);這6組的組織PDCD4分別為(101.06±2.49),(31.12±1.37),(35.25±1.13),(79.62±1.73),(79.52±1.45),(56.40±2.82)ng·L~(-1);這6組的血清PDCD4分別為(22.00±1.56),(6.93±0.62),(7.78±0.67),(19.92±1.01),(19.13±4.42),(15.34±1.02)ng·L~(-1)。與對(duì)照組相比,中高2個(gè)劑量實(shí)驗(yàn)組與模型組血清E2、存活素、組織Ki67與P53水平降低,差異有統(tǒng)計(jì)學(xué)意義(均P0.05);與模型組比較,中高2個(gè)劑量實(shí)驗(yàn)組的血清E2、存活素、組織Ki67與P53水平較低,差異有統(tǒng)計(jì)學(xué)意義(均P0.05)。與對(duì)照組比較,中高2個(gè)劑量實(shí)驗(yàn)組與模型組組織與血清PDCD4水平較高,差異有統(tǒng)計(jì)學(xué)意義(P0.05);與模型組比較,中高2個(gè)劑量實(shí)驗(yàn)組的組織與血清PDCD4水平較高,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論復(fù)方苦參注射液能夠明顯改善乳腺癌模型大鼠Ki67、存活素、P53及PDCD4水平。
[Abstract]:Objective to study the effect of compound Sophora flavescens injection on breast cancer model rats. Methods Fifteen male SD rats were selected as normal group according to random number method. The breast cancer model was induced by intraperitoneal injection of (MNU). The successful rats were randomly divided into 5 groups: model group, control group, three dose experimental groups, each with 15 rats. From the second day after the successful establishment of the model, the experimental group was intraperitoneally injected with compound Sophora flavescens injection 80 (40g / ml) kg~ (-1), the model group was administrated with 40 mL kg~ (-1) of tamoxifen suspension, and the rats in the control group and the normal group were given 4 mL kg~ (-1) intragastrically, once a day in each group. After 9 weeks of intervention, the levels of estradiol (E2) and survivin in serum, cell proliferation antigen marker (Ki67) in mammary gland, p53 gene p53 and programmed cell death factor 4 (PDCD4) in tissue and serum were measured by enzyme-linked immunosorbent assay (Elisa).
