【摘要】：目的探究肝纖溶顆粒與復(fù)方鱉甲軟肝片對(duì)大鼠肝纖維化模型細(xì)胞凋亡及內(nèi)質(zhì)網(wǎng)應(yīng)激相關(guān)因子表達(dá)影響。方法 48只雄性清潔級(jí)SD大鼠隨機(jī)分為:正常對(duì)照組、模型對(duì)照組、復(fù)方鱉甲軟肝片組及肝纖溶顆粒組,每組各12只。除正常對(duì)照組外各組復(fù)制肝纖維化模型。于復(fù)制模型后的3周各組大鼠同時(shí)給藥,正常對(duì)照組與模型對(duì)照組大鼠給予生理鹽水1.5 ml/kg灌胃,1次/d;復(fù)方鱉甲軟肝片組給予33.3%復(fù)方鱉甲軟肝片溶液1.5 ml/kg重灌胃,1次/d;肝纖溶顆粒組給予33.3%肝纖溶顆粒溶液1.5 ml/kg灌胃,1次/d。各組連續(xù)用藥6周。實(shí)驗(yàn)開始后第3、6和9周對(duì)各組大鼠肝臟病理學(xué)、細(xì)胞凋亡數(shù)量、肝纖維化指標(biāo)水平以及內(nèi)質(zhì)網(wǎng)應(yīng)激相關(guān)指標(biāo)進(jìn)行檢測(cè)。結(jié)果與其他各組相比,肝纖溶顆粒組纖維增生程度評(píng)分下降(P0.05),肝凋亡數(shù)下降(P0.05);透明質(zhì)酸、層黏連蛋白、血清Ⅲ型前膠原、血清Ⅳ型膠原水平下降(P0.05);CHOP、ATF6蛋白表達(dá)水平下降(P0.05)。結(jié)論肝纖溶顆粒組肝纖維化大鼠細(xì)胞凋亡數(shù)量和肝纖維化水平較復(fù)方鱉甲軟肝片組下降,差異有統(tǒng)計(jì)學(xué)意義(P0.05),肝纖溶顆粒能夠改善肝臟病理變化,調(diào)節(jié)內(nèi)質(zhì)網(wǎng)應(yīng)激相關(guān)指標(biāo),對(duì)臨床有指導(dǎo)意義。
[Abstract]:Objective to investigate the effects of Ganxian granule and compound Biejia Ruangan tablet on apoptosis and expression of ER stress related factors in rat liver fibrosis model. Methods Forty-eight male SD rats of clean grade were randomly divided into normal control group, model control group, compound Biejia Ruanggan tablet group and Ganxian granule group with 12 rats in each group. Hepatic fibrosis model was established in each group except the normal control group. Rats in each group were given drugs at the same time at 3 weeks after the establishment of the model. Normal control group and model control group were given normal saline 1.5 ml/kg intragastric perfusion once a day, compound Biejia Ruanggan tablet group was given 33.3% compound Biejia Ruanggan tablet solution 1.5 ml/kg reperfused once a day, and liver fibrinolytic granule group was given 33.3% liver fibrinolytic granule solution 1.5 ml/kg intragastrically, once a day. Each group was treated continuously for 6 weeks. Liver pathology, apoptosis, liver fibrosis and endoplasmic reticulum stress were measured at the 6th and 9th week after the experiment.
