【摘要】：目的:探討表皮生長因子(EGF)對中年大鼠運動活性以及紋狀體多巴胺含量的影響。方法:經(jīng)皮給予健康18月齡大鼠不同劑量EGF(0.01,0.05,0.1,0.15,0.3 mg/ml),通過曠場實驗檢測大鼠運動行為的改變,通過液相色譜-串聯(lián)質(zhì)譜分析多巴胺的含量變化。結(jié)果:與對照組相比,小劑量EGF組大鼠(0.01,0.05 mg/ml)曠場實驗中各行為參數(shù)沒有變化;中等劑量EGF組大鼠(0.1,0.15 mg/ml)曠場實驗中聞嗅行為、探索行為、趨觸性行為、運動行為和理毛行為改善;大劑量EGF(0.3 mg/m1)組大鼠曠場實驗中各行為參數(shù)下降。紋狀體多巴胺及其代謝產(chǎn)物DOPAC和HVA的含量與對照組相比,小劑量EGF(0.01 mg/rnl)組開始上升,中等劑量EGF(0.1 mg/ml)組達到峰值,大劑量EGF(0.3 mg/ml)組下降。結(jié)論:EGF對中年大鼠運動活性以及紋狀體多巴胺的含量均有影響。
[Abstract]:Aim: to investigate the effects of epidermal growth factor (EGF) on motor activity and dopamine content in striatum of middle-aged rats. Methods: healthy 18-month-old rats were given different doses of EGF (0.010.05U 0.150.30 mg/ml). The changes of exercise behavior and dopamine content were detected by open-field experiment and HPLC / tandem mass spectrometry respectively. The changes of dopamine content in rats were analyzed by HPLC and tandem mass spectrometry (LC-MS). Results: compared with the control group, there was no change in the behavioral parameters in the open field test of the rats in the low dose EGF group (0. 01 鹵0. 05 mg/ml), while in the middle dose EGF group (0. 1 0. 15 mg/ml), the sniffing behavior, exploratory behavior, contact behavior, motor behavior and hairdressing behavior of the rats in the middle dose EGF group (0. 1 0. 15 mg/ml) were improved. The behavioral parameters of large dose EGF (0. 3 mg/m1) group were decreased in open field experiment. Compared with the control group, the contents of dopamine and its metabolites DOPAC and HVA in striatum began to increase in the low dose EGF (0. 01 mg/rnl) group, reached the peak in the middle dose EGF (0. 1 mg/ml) group, and decreased in the high dose EGF (0. 3 mg/ml) group. Conclusion: EGF has an effect on motor activity and dopamine content in striatum of middle-aged rats.
