中藥川貝母對(duì)哮喘模型小鼠MMP-2,MMP-9和TIMP-1的影響
[Abstract]:Objective: to observe the effects of Chinese traditional Chinese medicine Chuanbei on airway remodeling, matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9) and tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) in asthmatic mice. Model group, high dose group, low dose group, positive control group. The asthmatic mice were established by using ovalbumin (OVA) in other groups except the normal group. After the establishment of the model, the high dose group and the low dose group were perfused with the dose of 18.0 ~ 9.0 mg kg-1, respectively; the positive control group was intraperitoneally injected with 0.5 mg kg-1 dexamethasone; the normal group and the model group were given the same amount of normal saline once a day for 28 days. The changes of airway reactivity, the changes of total cell count and leukocyte classification in bronchoalveolar lavage fluid (BALF) and the thickness of bronchial wall and bronchial smooth muscle were observed. The level of MMP-2MMP 9 and TIMP-1 was detected by Elisa and the expression of TIMP-1 mRNA in MMP-2 MMP-9 and TIMP-1 was detected by RT-PCR. The results showed that the total number of cells in airway reactive BALF and the counts of neutrophils, macrophages, lymphocytes and eosinophilic granulocytes in the model group. The thickness of bronchial wall and the thickness of bronchial smooth muscle were significantly higher than that of normal group (P0.01), the levels of MMP-2 MMP-9 and TIMP-1 and the expression of mRNA in model group were significantly higher than those in normal group (P0.01), while those in high dose group, low dose group and positive control group were significantly lower than those in model group (P0.05 or P0.01). The high dose group, low dose group and positive control group were significantly lower than the model group. The difference was statistically significant (P0.01). It is inferred that Chuanbei can improve airway remodeling in asthmatic mice, and its mechanism may be related to the decrease of MMP-2, MMP-9 and TIMP-1.
