胃得安片對(duì)應(yīng)激性胃損傷模型大鼠胃功能及胃組織中EGF、EGFR表達(dá)的影響

【摘要】:目的:研究胃得安片對(duì)應(yīng)激性胃損傷模型大鼠胃功能及胃組織中表皮生長因子(EGF)及其受體(EGFR)表達(dá)的影響,探討其治療胃潰瘍的作用機(jī)制。方法:將大鼠隨機(jī)分為正常組(生理鹽水)、模型組(生理鹽水)、陽性組(鹽酸雷尼替丁,0.015 mg/kg)和胃得安片高、中、低劑量組(1.7、0.87、0.43 g/kg),每組10只。除正常組外,其余各組大鼠均采用空腹冷水游泳法復(fù)制胃潰瘍模型。成模后,各組大鼠每天ig給藥1次,連續(xù)2周。記錄大鼠于給藥第0、7、14天時(shí)的體質(zhì)量;末次給藥12 h后,檢測大鼠的胃液分泌量、胃液pH值、胃潰瘍面積及胃黏膜損傷指數(shù),并檢測胃組織中EGF、EGFR的表達(dá)。結(jié)果:與正常組比較,模型組大鼠給藥第7、14天時(shí)的體質(zhì)量及胃液pH值降低,胃液分泌量和胃潰瘍面積增加,胃黏膜損傷指數(shù)升高,胃組織中EGF、EGFR表達(dá)增強(qiáng),差異均有統(tǒng)計(jì)學(xué)意義(P0.05或P0.01)。與模型組比較,除胃得安片中劑量組大鼠給藥第7天時(shí)的體質(zhì)量、胃液分泌量、胃液pH值、胃潰瘍面積以及胃得安片低劑量組大鼠給藥第7、14天時(shí)的體質(zhì)量、胃液分泌量、胃液pH值、胃潰瘍面積改善和胃組織中EGF表達(dá)增強(qiáng)不顯著外,其余各給藥組大鼠上述指標(biāo)均顯著改善,且胃組織中EGF、EGFR表達(dá)持續(xù)增強(qiáng)(P0.05或P0.01);胃得安片的作用呈現(xiàn)一定的量效關(guān)系。結(jié)論:胃得安片能夠顯著改善應(yīng)激性胃損傷模型大鼠的胃功能;其機(jī)制可能與增強(qiáng)胃組織中EGF、EGFR表達(dá),從而促進(jìn)潰瘍愈合有關(guān)。
[Abstract]:Aim: to study the effects of Weidian tablets on gastric function and the expression of epidermal growth factor (EGF) and epidermal growth factor receptor (EGFR) in gastric tissues of stress induced gastric injury rats, and to explore the mechanism of its action in the treatment of gastric ulcer. Methods: the rats were randomly divided into normal group (normal saline), model group (normal saline), positive group (Ranitidine hydrochloride 0.015 mg/kg) and high, middle and low dose group (1.70.87 0.43 g/kg) with 10 rats in each group. Gastric ulcer was induced by cold water swimming in all groups except the normal group. Rats in each group were given intravenously once a day for 2 weeks. The body weight of the rats was recorded at 714 days after the last administration, and the gastric juice secretion, gastric pH value, gastric ulcer area and gastric mucosal injury index were measured 12 hours after the last administration, and the expression of EGF- EGFR in gastric tissue was detected. Results: compared with the normal group, the body weight and pH value of gastric juice decreased, the gastric juice secretion and gastric ulcer area increased, the gastric mucosal injury index increased, and the EGF- EGFR expression in gastric tissue increased in the model group on the 14th day after administration of the drug. The difference was statistically significant (P0.05 or P0.01). Compared with the model group, the body weight, gastric juice secretion, gastric juice pH value, gastric ulcer area, the body weight and gastric juice secretion of the rats in the middle dose group at 714 days after administration were compared with those in the model group. The pH value of gastric juice, the area of gastric ulcer and the expression of EGF in gastric tissue were not significantly improved, but the above indexes were significantly improved in the other groups (P0.05 or P0.01), and the effect of Wedelan tablets was dose-effect. Conclusion: Weidian tablet can significantly improve gastric function in rats with stress gastric injury, and its mechanism may be related to the enhancement of EGF- EGFR expression in gastric tissue and thus to promote ulcer healing.
