【摘要】：為了評價益生菌制劑對腹瀉模型小鼠腸道菌群的恢復(fù),現(xiàn)采用22.4 g/(kg·d)氨芐青霉素灌胃構(gòu)造小鼠腹瀉模型,其中空白對照組灌胃等量生理鹽水。造模成功后,用低、中、高不同劑量的益生菌制劑對小鼠治療恢復(fù),治療14 d后,觀察益生菌制劑對小鼠稀便率、稀便級和腹瀉指數(shù)的影響。無菌取治療后0、7、14 d的小鼠糞便進行分析,分別采用平板計數(shù)和DGGE技術(shù)檢測腸道目標(biāo)菌群變化。停用益生菌制劑后,無菌采取0、7、14 d的糞便樣品,熒光定量PCR定量檢測外源益生菌。結(jié)果表明,益生菌制劑不僅能改善小鼠的腹瀉情況,促進腹瀉小鼠的生長,還對腹瀉小鼠腸道菌群結(jié)構(gòu)具有顯著恢復(fù)作用;熒光定量PCR檢測結(jié)果表明外源益生菌嗜酸乳桿菌NCFM、乳雙歧桿菌Bi-07和鼠李糖乳桿菌HN001在停止灌胃益生菌制劑后可定殖存活,但隨著時間推移呈一定衰減的趨勢。
[Abstract]:In order to evaluate the effect of probiotics on the recovery of intestinal flora in mice with diarrhea, the diarrhea model was established by gastric administration of 22.4 g / (kg d) ampicillin, in which the blank control group was given the same amount of normal saline. After successful modeling, the mice were treated with low, medium and high doses of probiotics. After 14 days of treatment, the effects of probiotics on dilute stool rate, dilute stool grade and diarrhea index were observed. The faeces of mice treated with aseptic therapy for 14 days were analyzed. Plate count and DGGE technique were used to detect the changes of intestinal flora. After the probiotics were stopped, the fecal samples of 0 ~ 7 ~ 14 days were used for quantitative detection of exogenous probiotics by fluorescence quantitative PCR. The results showed that probiotics could not only improve the diarrhea of mice and promote the growth of diarrhea mice, but also significantly restore the intestinal flora structure of diarrhea mice. Fluorescence quantitative PCR analysis showed that exogenous probiotics, Lactobacillus acidophilus NCFM, Lactobacillus acidophilus Bi-07 and Lactobacillus rhamnoides HN001 could colonize and survive after stopping gastric administration of probiotics, but tended to decrease with time.
