腺病毒7d基因型與7b基因型全基因組比較分析

【摘要】:為了解重慶地區(qū)流行的7型腺病毒(Human adenovirus 7,HAdV-7)基因型型別,從全基因組水平探討HAdV-7b和HAdV-7d可能的致病性差異,收集兒童急性下呼吸道感染患兒的呼吸道分泌物標(biāo)本分離培養(yǎng),提取DNA后進(jìn)行HAdV-7基因型別鑒定、限制性酶切分析和全基因組測(cè)序,再利用MEGA6比較分析。本研究在2009年6月~2013年1月間共收集4334份急性下呼吸道感染患兒的鼻咽抽吸物標(biāo)本,隨機(jī)選取2411份進(jìn)行病毒分離培養(yǎng),共有164份(164/2411,6.8%)鼻咽抽吸物HAdV陽(yáng)性,其中HAdV-7 87例。限制性酶切分析顯示,隨機(jī)挑選的3株HAdV-7分離株均為HAdV-7d。CQ1198株行全基因組測(cè)序并與其他HAdV-7全基因組構(gòu)建進(jìn)化樹,結(jié)果發(fā)現(xiàn)16株中國(guó)地區(qū)的HAdV-7為HAdV-7d基因型,且上述16株HAdV-7與7d2的同源性為99.9%,與7b的同源性為98.2%。相較于ak40株(HAdV-7b),CQ1198共有49處核苷酸替換,其中20處可導(dǎo)致非同義替換;4處核苷酸插入,其中24bp和18bp 2個(gè)片段插入分別位于病毒相關(guān)RNAⅡ(Virus-associated RNAⅡ,VA RNAⅡ)和L3的pⅥ區(qū)域。HAdV-7可有多個(gè)基因型致病,且一直處于進(jìn)化過(guò)程中,其主要流行基因型由HAdV-7b變?yōu)镠AdV-7d。應(yīng)加強(qiáng)對(duì)HAdV分子流行病學(xué)監(jiān)測(cè),明確其進(jìn)化規(guī)律,為HAdV疫苗研發(fā)提供分子流行病學(xué)依據(jù)。
[Abstract]:In order to understand the genotypes of adenovirus type 7 (Human adenovirus 7) HAdV-7, the possible pathogenicity differences between HAdV-7b and HAdV-7d were studied at the whole genome level. The samples of respiratory secretions from children with acute lower respiratory infection were collected and cultured. HAdV-7 genotyping, restriction enzyme digestion and whole genome sequencing were performed after DNA extraction, and then MEGA6 was used to compare and analyze. From June 2009 to January 2013, a total of 4334 nasopharyngeal aspiration samples were collected from children with acute lower respiratory tract infection. 2411 samples were isolated and cultured randomly. A total of 164 cases (164 / 24116.8%) were HAdV positive in nasopharynx aspiration, among which 87 cases were HAdV-7. Restriction endonuclease analysis showed that all of the three randomly selected HAdV-7 isolates were HAdV-7d.CQ1198 strains and constructed phylogenetic tree with other HAdV-7 genomes. The results showed that 16 HAdV-7 strains in China were HAdV-7d genotypes. The homology between HAdV-7 and 7d2 was 99.9 and 98.2 with 7b.
