氯化鋰對(duì)脊髓損傷模型大鼠運(yùn)動(dòng)功能的作用和機(jī)制

【摘要】:[目的]觀察氯化鋰對(duì)脊髓損傷后BDNF/TrkB信號(hào)表達(dá)的作用,探討其對(duì)脊髓損傷大鼠肢體運(yùn)動(dòng)功能恢復(fù)的作用和機(jī)制。[方法]36只SD大鼠隨機(jī)分為假手術(shù)組(12例)、對(duì)照組(12例)、氯化鋰組(12例),假手術(shù)組僅行椎板切除術(shù),對(duì)照組和氯化鋰組采用NYU脊髓打擊器建立脊髓損傷模型,術(shù)后第1、3、5 d和第7 d行BBB肢體運(yùn)動(dòng)功能評(píng)分。干預(yù)7 d后取材,采用尼氏染色光鏡觀察神經(jīng)元形態(tài),免疫熒光定位BDNF陽性細(xì)胞,并計(jì)數(shù)進(jìn)行半定量分析,Western blot檢測(cè)BDNF蛋白、TrkB蛋白和p-TrkB蛋白表達(dá)。[結(jié)果]在脊髓損傷后肢體運(yùn)動(dòng)功能方面,自干預(yù)后第3 d起,氯化鋰組BBB評(píng)分高于對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(P0.05),至術(shù)后第5 d,兩組間BBB評(píng)分差異更加明顯(P0.01)。尼氏染色提示氯化鋰組脊髓損傷后神經(jīng)元的形態(tài)優(yōu)于對(duì)照組。氯化鋰組脊髓損傷后脊髓前角運(yùn)動(dòng)神經(jīng)元BDNF陽性細(xì)胞數(shù)高于對(duì)照組,差異具有統(tǒng)計(jì)學(xué)意義(P0.01);Western blot檢測(cè)表明氯化鋰組的BDNF蛋白和p-TrkB蛋白表達(dá)高于對(duì)照組,兩組間差異有統(tǒng)計(jì)學(xué)意義(P0.05),但兩組間TrkB蛋白表達(dá)差異無統(tǒng)計(jì)學(xué)意義(P0.05)。[結(jié)論]氯化鋰能促進(jìn)脊髓損傷對(duì)照大鼠肢體運(yùn)動(dòng)功能恢復(fù),其機(jī)制可能與其促進(jìn)脊髓損傷后脊髓前角運(yùn)動(dòng)神經(jīng)元分泌BDNF,促進(jìn)TrkB受體磷酸化,從而上調(diào)BDNF/TrkB信號(hào)表達(dá),促進(jìn)脊髓損傷后神經(jīng)元存活、再生和軸突再生、再髓鞘化有關(guān)。
[Abstract]:[objective] to observe the effect of lithium chloride on the expression of BDNF/TrkB signal after spinal cord injury (sci), and to explore its effect and mechanism on the recovery of limb motor function in rats with spinal cord injury (sci). [methods] Thirty-six SD rats were randomly divided into three groups: sham operation group (n = 12), control group (n = 12) and lithium chloride group (n = 12). The motor function of limbs was evaluated with BBB on the 1st and 7th day after operation. After 7 days of intervention, the morphology of neurons was observed with Nissl staining light microscope, the BDNF positive cells were localized by immunofluorescence, and the expression of BDNF protein TrkB protein and p-TrkB protein were detected by semi-quantitative analysis by Western blot. [results] from the 3rd day after the intervention, the BBB score of the lithium chloride group was higher than that of the control group (P0.05), and the difference between the two groups was more obvious than that of the control group (P0.01). Nissl staining showed that the morphology of neurons after spinal cord injury in lithium chloride group was superior to that in control group. The number of BDNF positive cells in spinal cord anterior horn motor neurons after spinal cord injury in lithium chloride group was higher than that in control group, and the difference was statistically significant (P0.01) Western blot analysis showed that the expression of BDNF protein and p-TrkB protein in lithium chloride group was higher than that in control group. The difference between the two groups was statistically significant (P0.05), but there was no significant difference in the expression of TrkB protein between the two groups (P0.05). [conclusion] Lithium chloride can promote the recovery of limb motor function in spinal cord injury control rats, and its mechanism may be similar to that of TrkB receptor phosphorylation in spinal cord anterior horn motoneurons after spinal cord injury, thereby up-regulating the expression of BDNF/TrkB signal. Promoting neuronal survival, regeneration and axonal regeneration after spinal cord injury is related to demyelination.
