【摘要】：目的探討瑞舒伐他汀鈣對老年膿毒癥大鼠急性肺損傷的干預(yù)作用。方法將72只老年雄性Wistar大鼠隨機分為假手術(shù)組、模型組、瑞舒伐他汀鈣干預(yù)組,每組24只。干預(yù)組及模型組采用改良盲腸結(jié)扎穿孔術(shù)方法建立老年膿毒癥ALI大鼠模型,假手術(shù)組建立假手術(shù)模型。術(shù)前1 w干預(yù)組開始每日給予瑞舒伐他汀鈣,劑量2.5 mg·kg~(-1)·d~(-1)。模型組和假手術(shù)組同一時間給予等量生理鹽水。實驗前24 h灌注藥物后禁食,自由飲水。每組大鼠平均分為3個亞組,分別于術(shù)后6、12和24 h取材。應(yīng)用酶聯(lián)免疫吸附法(ELISA)檢測血清細胞因子白細胞介素(IL)-2、IL-6、腫瘤壞死因子(TNF)-α水平、檢測動脈血氧分壓(PaO_2)并觀察肺組織的病理改變。結(jié)果模型組PaO_2明顯低于假手術(shù)組和干預(yù)組(P0.05),且隨時間延長而降低。模型組血清細胞因子IL-2、IL-6、TNF-α水平高于假手術(shù)組和干預(yù)組(P0.05),與模型組比,干預(yù)組肺損傷程度較輕。結(jié)論瑞舒伐他汀鈣能抑制老年膿毒癥大鼠ALI的炎癥反應(yīng)、改善氧分壓,進而減輕ALI。
[Abstract]:Objective to investigate the effect of rosuvastatin calcium on acute lung injury in aged sepsis rats. Methods Seventy-two aged male Wistar rats were randomly divided into sham-operation group, model group and rosuvastatin calcium intervention group with 24 rats in each group. The intervention group and the model group established the model of senile sepsis Ali by modified cecal ligation and perforation, and the sham operation group established the sham-operation model. The intervention group was given recuvastatin calcium daily at a dose of 2.5 mg kg ~ (-1) d ~ (-1). The model group and the sham operation group were given the same amount of saline at the same time. The drug was infused 24 hours before the experiment and fasting and drinking water was free. The rats in each group were divided into 3 subgroups on average. The samples were obtained at 6 h and 24 h postoperatively. The levels of serum interleukin (IL)-2 and tumor necrosis factor-偽 (TNF-偽) were measured by enzyme linked immunosorbent assay (Elisa). The arterial partial pressure of oxygen (Pao) was measured and the pathological changes of lung tissue were observed. Results PaO2 in model group was significantly lower than that in sham operation group and intervention group (P0.05), and decreased with time. The level of serum cytokine IL-6 TNF- 偽 in the model group was higher than that in the sham operation group and the intervention group (P0.05). Compared with the model group, the degree of lung injury in the intervention group was less than that in the control group. Conclusion Risuvastatin calcium can inhibit the inflammatory response of Ali in aged sepsis rats, improve the partial pressure of oxygen, and then alleviate the ALI.
