菖蒲郁金湯對(duì)多發(fā)性抽動(dòng)癥模型大鼠單胺類神經(jīng)遞質(zhì)的影響

【摘要】:目的:觀察菖蒲郁金湯對(duì)多發(fā)性抽動(dòng)癥(TS)模型大鼠單胺類神經(jīng)遞質(zhì)的影響,探討其治療多發(fā)性抽動(dòng)癥的作用機(jī)理。方法:72只雄性SD大鼠隨機(jī)分為空白組12只,造模組60只,造模組大鼠腹腔注射亞氨基二丙腈(IDPN)制備多發(fā)性抽動(dòng)癥動(dòng)物模型,空白組大鼠腹腔注射等體積的生理鹽水,每天1次,連續(xù)7天,造模完成后,造模組大鼠表現(xiàn)出不同程度的運(yùn)動(dòng)行為及刻板行為,評(píng)分均≥2分,表明造模成功。造模成功后,將造模組大鼠再次分為:模型組、泰必利組、菖蒲郁金湯77.28g/kg、38.64g/kg、19.32g/kg組,各12只。從造模完第2天開始給予灌胃,泰必利組給予泰必利(3.194mg/ml)混懸液,菖蒲郁金湯77.28g/kg、38.64g/kg、19.32g/kg組分別給予相應(yīng)濃度的菖蒲郁金湯(5.152、2.576、1.288g生藥/ml)藥液,空白組與模型組給予等體積純凈水,每天1次,連續(xù)給藥4周。在最后一次給藥后24 h采集標(biāo)本,用ELISA法檢測(cè)大鼠腦組織及血漿多巴胺(DA)、去甲腎上腺素(NE)、5-羥色胺(5-HT)及高香草酸(HVA)水平。結(jié)果:(1)模型組大鼠紋狀體DA含量明顯下降,腦干5-HT含量明顯升高;治療后泰必利組大鼠紋狀體DA含量明顯升高,菖蒲郁金湯77.28g/kg組大鼠紋狀體DA、HVA含量及腦干5-HT含量明顯升高。(2)模型組大鼠血漿HVA含量明顯降低;治療后,泰必利組大鼠血漿HVA含量明顯升高,菖蒲郁金湯77.28g/kg、38.64g/kg組大鼠血漿HVA含量明顯升高,菖蒲郁金湯77.28g/kg、38.64g/kg、19.32g/kg組大鼠血漿5-HT含量明顯升高。結(jié)論:單胺類神經(jīng)遞質(zhì)間的平衡失調(diào)可能與多發(fā)性抽動(dòng)癥發(fā)病有關(guān),而菖蒲郁金湯可能具有調(diào)節(jié)單胺類神經(jīng)遞質(zhì)間平衡的作用。
[Abstract]:Objective: to observe the effect of Aulubuyujin decoction on monoamine neurotransmitters in rats with multiple tic syndrome (TS) and to explore its mechanism. Methods Seventy-two male Sprague-Dawley rats were randomly divided into blank group (n = 12) and model group (n = 60). Rats in the model group were treated with intraperitoneal injection of iminodipropionitrile (IDPN) to make the animal model of multiple tic syndrome. Rats in the blank group were given intraperitoneal injection of normal saline of the same volume once a day.
