【摘要】：目的觀察平?jīng)_降逆方對裸鼠人子宮內(nèi)膜異位癥(endometriosis,EMs)模型種植灶凋亡、TNF-α、IL-8表達的影響。方法將48只BALB/c-nu雌性裸鼠,隨機分為兩組:EMs源性模型組和非EMs源性模型組,每組24只。EMs源性模型組采用EMs患者的子宮內(nèi)膜種植模型鼠;非EMs源性模型組采用非EMs患者的子宮內(nèi)膜種植模型鼠。兩組再分別隨機分為中藥組、陽性藥物組和陰性對照組,每組8只。分別給予平?jīng)_降逆方、米非司酮及等量生理鹽水灌胃給藥4周。觀察模型鼠種植灶組織學形態(tài)及超微結(jié)構(gòu)、細胞凋亡、TNF-α、IL-8表達。結(jié)果 EMs及非EMs模型鼠的中藥組均見病灶縮小、周圍血管生成少于同源陰性對照組,光鏡、電鏡下可見內(nèi)膜組織退變,有凋亡現(xiàn)象發(fā)生,免疫組化結(jié)果示凋亡率明顯高于同源陰性對照組,而TNF-α、IL-8表達量明顯低于同源陰性對照組(P0.05)。結(jié)論平?jīng)_降逆方可明顯抑制裸鼠異位內(nèi)膜的生長,這種作用可能是通過誘導細胞凋亡、降低TNF-α、IL-8表達而實現(xiàn)的。
[Abstract]:Objective To observe the effect of Ping Chong and descending Recipe on the apoptosis, TNF- alpha and IL-8 expression of endometriosis (EMs) model in nude mice. Methods 48 BALB/c-nu female nude mice were randomly divided into two groups: EMs source model group and non EMs model group, 24.EMs derived model groups in each group adopted the endometrial implant model of EMs patients. The non EMs source model group used the non EMs endometrium model rats. The two groups were randomly divided into the Chinese medicine group, the positive drug group and the negative control group, with 8 rats in each group. The group was given the square flushing and the descending side, mifepristone and the same amount of normal saline for 4 weeks. Death, TNF- alpha, IL-8 expression. Results both EMs and non EMs model rats were found to reduce the lesion and the peripheral blood vessels were less than the homologous negative control group. Light microscopy and electron microscopy showed that the intima tissue degeneration and apoptosis occurred. The immunohistochemical results showed that the apoptosis rate was significantly higher than that of the homologous negative control group, and the expression of TNF- alpha and IL-8 was significantly lower than that of the homologous negative group. Sex control group (P0.05). Conclusion Ping Chong and descending recipe can obviously inhibit the growth of ectopic endometrium in nude mice. This effect may be achieved by inducing apoptosis and reducing the expression of TNF- alpha and IL-8.
