【摘要】:目的探討犬骨髓基質(zhì)細(xì)胞(bone marrow stromal cells, BMSCs)細(xì)胞片層在構(gòu)建組織工程骨中的作用。 方法制備犬同種異體脫鈣骨基質(zhì)(dermineralized bone matrix, DBM)。將人重組骨形態(tài)發(fā)生蛋白-2(recombination human bone morphogenetic protein-2, rhBMP-2)復(fù)合到DBM上。抽取犬髂骨骨髓,采用密度梯度離心法分離犬骨髓基質(zhì)細(xì)胞(BMSCs),將其向成骨細(xì)胞誘導(dǎo)分化。將經(jīng)成骨誘導(dǎo)的第3代細(xì)胞接種于溫度反應(yīng)性培養(yǎng)皿中培養(yǎng),至細(xì)胞增殖并互相融合時(shí),將培養(yǎng)箱溫度降至20℃,制備完整的BMSCs細(xì)胞片層,將其包裹DBM/rhBMP-2/BMSCs復(fù)合體植入犬左側(cè)背闊肌下包裹胸背動(dòng)靜脈束為實(shí)驗(yàn)側(cè),以無(wú)BMSCs細(xì)胞片層包裹的DBM/rhBMP-2/BMSCs復(fù)合體植入犬右側(cè)背闊肌下包裹胸背動(dòng)靜脈束為對(duì)照側(cè)。術(shù)后8、12、16周取材行大體觀察、影像學(xué)觀察及組織學(xué)觀察,評(píng)價(jià)成骨情況。采用SPSS18.0統(tǒng)計(jì)軟件對(duì)數(shù)據(jù)進(jìn)行兩樣本均數(shù)差別的t檢驗(yàn)。 結(jié)果 1.制備的BMSCs細(xì)胞片層完整致密,在支架復(fù)合體表面生長(zhǎng)良好。 2.成骨面積實(shí)驗(yàn)側(cè)對(duì)照側(cè),兩者之間差異有顯著性(P0.05)。術(shù)后16周,實(shí)驗(yàn)側(cè)有大量板層骨形成,有哈弗氏系統(tǒng)形成,骨髓腔內(nèi)有紅骨髓;對(duì)照側(cè)有板層骨形成,無(wú)哈弗氏系統(tǒng)形成,骨髓腔內(nèi)無(wú)紅骨髓。 結(jié)論BMSCs細(xì)胞片層可促進(jìn)具有致密板層骨和哈弗氏系統(tǒng)的組織工程骨的形成。
[Abstract]:Objective to investigate the role of bone marrow stromal cells, BMSCs) cell lamellae in the construction of tissue engineered bone in canine bone marrow stromal cells. Methods canine allogeneic demineralized bone matrix (DBM) was prepared. Human recombinant bone morphogenetic protein-2 recombination human bone morphogenetic protein-2 (rhBMP-2) was synthesized into DBM. The bone marrow of iliac bone of dogs was extracted and the bone marrow stromal cells (BMSCs) were isolated by density gradient centrifugation and differentiated into osteoblasts. The third passage cells induced by osteogenesis were cultured in a temperature-reactive culture dish. When the cells proliferated and fused with each other, the incubator temperature was lowered to 20 鈩,
