腦出血免疫抑制大鼠模型的構建及評估

【摘要】:背景:研究表明,大鼠腦出血后經治療可以有效抑制其免疫功能。已有文獻研究對缺血性腦卒中后免疫抑制現(xiàn)象進行詳細研究,但腦出血后機體免疫功能研究鮮見報道。目的:構建腦出血后免疫抑制大鼠模型并進行穩(wěn)定性評估。方法:將60只Wistar大鼠隨機分為3組,即對照組、假手術組、腦出血組,每組20只。采用自身動脈血50μL注入大鼠基底節(jié)方法建造急性腦出血模型;假手術模型注入50μL生理鹽水,手術過程同腦出血模型;對照組不做任何處理。分別于造模后24,48,72,96 h用血液分析儀檢測白細胞、淋巴細胞、淋巴細胞百分數(shù);采用ELISA測定大鼠血清中促炎細胞因子白細胞介素6和抗炎細胞因子轉化生長因子β的水平;剖取大鼠脾組織進行組織學及病理學檢查;通過RT-PCR及Western blotting檢測轉化生長因子β、白細胞介素6基因和蛋白在脾組織中的表達變化。結果與結論:(1)與假手術組及對照組相比,腦出血組大鼠于造模后24,48,72,96 h白細胞數(shù)明顯升高,淋巴細胞百分數(shù)逐漸降低,差異有顯著性意義(P0.05);(2)與假手術組及對照組相比,腦出血組大鼠血液、脾的白細胞介素6在24 h時高表達,72 h達高峰,96 h時表達降低,差異有顯著性意義(P0.05);(3)與假手術組及對照組相比,腦出血組大鼠血液、脾的轉化生長因子β于24 h時低表達,72 h逐漸升高,96 h時高表達,差異有顯著性意義(P0.05);(4)結果提示,大鼠在腦出血后先呈現(xiàn)免疫功能亢奮,隨即96 h后出現(xiàn)免疫功能抑制,說明腦出血后免疫抑制大鼠模型造模成功,并具有良好的穩(wěn)定性。
[Abstract]:Background: studies have shown that intracerebral hemorrhage after treatment can effectively inhibit its immune function. The phenomenon of immunosuppression after ischemic stroke has been studied in detail, but the study of immune function after cerebral hemorrhage is seldom reported. Objective: to establish an immunosuppressive rat model after intracerebral hemorrhage and evaluate its stability. Methods: 60 Wistar rats were randomly divided into 3 groups: control group, sham operation group and intracerebral hemorrhage group. The acute intracerebral hemorrhage model was established by injecting 50 渭 L autogenous arterial blood into the basal ganglia of rats, and the sham operation model was injected with 50 渭 L normal saline. The percentage of leukocytes and lymphocytes were detected by blood analyzer at 24: 48 and 72 ~ 96 hours after modeling, and the levels of pro-inflammatory cytokine interleukin-6 and anti-inflammatory cytokine transforming growth factor 尾 in serum of rats were measured by ELISA. The changes of expression of transforming growth factor 尾, interleukin 6 gene and protein in spleen tissue were detected by RT-PCR and Western blotting. Results compared with the sham operation group and the control group, the leukocyte count and the lymphocyte percentage in the cerebral hemorrhage group were significantly increased and the lymphocyte percentage was gradually decreased in the cerebral hemorrhage group compared with the sham operation group and the control group (P < 0.05), compared with the sham operation group and the control group. Compared with the sham operation group and the control group, the high expression of interleukin 6 in the spleen decreased at 72 h and reached the peak at 96 h after ICH, and the difference was significant (P 0.05) compared with the sham operation group and the control group, the blood of the ICH group was significantly lower than that of the sham operation group and the control group. The low expression of TGF- 尾 in spleen increased gradually at 72 h after 24 h and increased gradually at 96 h. The difference was significant (P 0.05). The results showed that the immune function of rats was hyperactive after cerebral hemorrhage, and then immunosuppression appeared 96 h later. The results showed that the model of immunosuppressive rats after intracerebral hemorrhage was successful and had good stability.
