【摘要】：研究豬帶絳蟲重組BCG-TSOL18疫苗免疫仔豬,豬帶絳蟲卵攻擊后誘導其保護性免疫應(yīng)答的情況。將12頭40d齡健康仔豬均分為3組:分別為1011 CFU重組BCG-TSOL18疫苗灌胃組、BCG對照組和PBS對照組。每2周免疫1次,共免疫2次。于末次免疫4周后用豬帶絳蟲卵進行攻擊感染,3月后剖殺仔豬,分離囊尾蚴,計算減蚴率;取前腔靜脈血,分離血清和制備外周血單個核細胞(peripheral blood monouclear cell,PBMC)。采用ELISA法檢測血清IgG、IgG1及IgG2a水平以及PBMC培養(yǎng)上清液中IL-2、IFN-γ、IL-4及IL-10的水平;MTT比色法檢測PBMC增殖水平。重組BCG-TSOL18疫苗免疫組豬帶絳蟲卵攻擊后誘導仔豬獲得79.44%的保護力。與BCG和PBS對照組比較,重組BCG-TSOL18疫苗組血清IgG、IgG2a水平顯著升高(t=13.026,P=0.000;t=13.253,P=0.000;t=7.284,P=0.000;t=8.453,P=0.000),IgG1水平顯著降低(t=-9.118,P=0.000;t=-10.372,P=0.000);抗原刺激下PBMC顯著增殖(t=14.929,P=0.000;t=14.665,P=0.000);PBMC培養(yǎng)上清液IL-2、IFN-γ水平顯著升高(t=9.768,P=0.000;t=10.496,P=0.000;t=22.504,P=0.000;t=23.204,P=0.000),IL-4和IL-10水平顯著降低(t=-18.659,P=0.000;t=-19.495,P=0.000;t=-26.258,P=0.000;t=-27.402,P=0.000)。豬帶絳蟲重組BCG-TSOL18疫苗可誘導仔豬產(chǎn)生Th1型的保護性免疫應(yīng)答,從而抵抗豬帶絳蟲卵的感染攻擊。
[Abstract]:To study the protective immune response induced by the recombinant BCG-TSOL18 vaccine of Taenia solium and the egg attack of Taenia solium, 12 40d age healthy piglets were divided into 3 groups: 1011 CFU recombinant BCG-TSOL18 vaccine group, BCG control group and PBS control group, 1 times every 2 weeks and 2 times in CO immunization. The pigs were used for 4 weeks after the last immunization. The eggs of the tapeworm were infected. After March, the piglets were killed, the cysticercus was separated and the rate of Echinococcus was calculated; the blood of the anterior vena cava, the serum and the peripheral blood mononuclear cells (peripheral blood monouclear cell, PBMC) were obtained. The level of IgG, IgG1 and IgG2a in the serum and IL-2, IFN- gamma, and the levels of PBMC were detected by ELISA. The proliferation level of PBMC was detected by colorimetric method. The recombinant BCG-TSOL18 Vaccine Immunized with Taenia solium eggs to induce 79.44% protection. Compared with the control group of BCG and PBS, the serum IgG and IgG2a level of the recombinant BCG-TSOL18 vaccine group increased significantly (t=13.026, P=0.000; t=13.253, P= 0; Low (t=-9.118, P=0.000; t=-10.372, P=0.000); PBMC proliferation under the stimulation of antigen (t=14.929, P=0.000; t=14.665, P=0.000); PBMC culture supernatant IL-2, IFN- gamma level significantly increased 8, P=0.000; t=-27.402, P=0.000). Recombinant BCG-TSOL18 vaccine of Taenia solium can induce Th1 type protective immune response in piglets, thus resisting the infection attack of Taenia solium eggs.
