骨髓間充質(zhì)干細(xì)胞聯(lián)合外源性Shh蛋白對(duì)骨髓造血干細(xì)胞增殖的影響

【摘要】:目的探討骨髓間充質(zhì)干細(xì)胞(BMMSC)聯(lián)合外源性Shh蛋白干預(yù)對(duì)骨髓造血干細(xì)胞(HSC)增殖的影響。方法分別采用全骨髓培養(yǎng)法、磁珠分選法(MASC)從正常志愿者骨髓中分離BMMSC、HSC,并用流式細(xì)胞術(shù)進(jìn)行鑒定。將分選好的HSC隨機(jī)分為三組,MSC+HSC+Shh組Transwell培養(yǎng)板上室接種用IMDM培養(yǎng)基+500ng/m L的Shh蛋白重懸的HSC,下室接種BMMSC;BMMSC+HSC組上室接種用IMDM培養(yǎng)基重懸后的HSC,下室接種BMMSC;HSC+Shh組上室接種用IMDM培養(yǎng)基+500 ng/mL的Shh蛋白重懸的HSC,下室加入培養(yǎng)基;對(duì)照組上室接種用IMDM培養(yǎng)基重懸的HSC,下室加入培養(yǎng)基。各組均培養(yǎng)72 h,用計(jì)數(shù)板計(jì)數(shù)HSC活細(xì)胞數(shù)。結(jié)果培養(yǎng)72 h,HSC+BMMSC+Shh組HSC細(xì)胞計(jì)數(shù)較其他各組高(P均0.05),HSC+MSC組、HSC+Shh組較HSC組高(P均0.05),HSC+BMMSC組、HSC+Shh組比較差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論 BMMSC與外源性Shh蛋白可協(xié)同作用共同促進(jìn)HSC增殖。
[Abstract]:Objective to investigate the effects of bone marrow mesenchymal stem cells (BMMSC) combined with exogenous Shh protein on the proliferation of bone marrow hematopoietic stem cells (HSCs). Methods BMMSC-HSCs were isolated from the bone marrow of normal volunteers by whole bone marrow culture method and magnetic bead sorting method respectively, and were identified by flow cytometry. The selected HSC were randomly divided into three groups: the Transwell HSC Shh group was inoculated with IMDM medium 500ng/m L Shh protein, the lower chamber group was inoculated with the IMDM medium, and the lower chamber group was inoculated with the IMDM medium 500ng/m L, and the lower chamber was inoculated with the upper chamber of the IMDM medium 500ng/m L, the lower chamber inoculated with the IMDM medium, and the lower chamber inoculated with the upper chamber of the BMMSC-HSC Shh group. The IMDM medium with 500 ng/mL Shh protein was inoculated and the lower chamber was added to the medium. In the control group, the upper chamber was inoculated with IMDM medium and the lower chamber was added to the medium. All groups were cultured for 72 hours and counted the number of HSC living cells with counter. Results the number of HSC cells in HSC BMMSC Shh group was higher than that in other groups (P < 0.05). There was no significant difference between HSC Shh group and HSC group (P 0.05). Conclusion BMMSC and exogenous Shh protein can promote the proliferation of HSC.
